为提高猪精液的保存效果,为猪精液常温保存技术及稀释液配方的研究提供参考依据,设计了3种常温保存希释液配方,并对保存后的精子进行了顶体染色。结果表明:当精子活率为50%与30%时,配方Ⅱ的精子保存时间均显著高于配方Ⅰ与配方Ⅲ(P〈0.05),且配方Ⅱ所保存精子的总存活时间也显著高于配方Ⅰ与配方Ⅲ(P〈0.05)。上述结果表明,配方Ⅱ对猪精液的常温保存效果要优于配方Ⅰ与配方Ⅲ。  相似文献

 为提高猪精液常温保存的效果,研究不同温度和稀释液pH对猪精液常温保存效果的影响。结果表明:(1)当精子活力分别为50%和30%时,配方Ⅰ与Ⅱ稀释液保存的精液,在温度为15~20 ℃条件下的精子存活时间均显著高于21~25 ℃(P<0.05);在pH为6.5~6.9条件下的精子存活时间均显著高于6.0~6.4(P<0.05);(2)精子顶体染色结果表明,适宜温度与pH条件下,精液保存48 h后,顶体完整率达到96%以上。研究结果表明,猪精液常温保存适宜温度为15~20 ℃,适宜PH为6.5~6.9。  相似文献

试验利用北京信邦稀释液(试验Ⅰ组)和台湾精旺稀释液(试验Ⅱ组)对猪精液进行常温保存,测定精子活率、有效存活时间、生存指数、pH值。试验结果:试验Ⅱ组稀释后第4天,精液的pH值显著高于试验Ⅰ组(P0.05),试验Ⅱ组的精液p H值始终高于试验Ⅰ组;试验Ⅱ组稀释后精子活率在第4天显著高于试验Ⅰ组(P0.05),试验Ⅱ组精子的有效存活时间与试验Ⅰ组差异不显著(P0.05)。试验结果表明,试验Ⅱ组采用的台湾精旺稀释液更有利于精液的保存,适宜大规模推广应用。  相似文献

为了研究五味子乙素(Sch B)对猪精液常温保存效果的影响,本实验设计6个猪精液常温保存稀释粉配方,在筛选出的最佳自配基础稀释粉配方中添加了0、1、5、10、15μmol/L的Sch B,通过检测精子活率、质膜完整性、顶体完整性以及总抗氧化能力(T-AOC)和丙二醛(MDA)含量,分析Sch B对猪精液常温保存效果的影响。结果表明:6号基础稀释粉猪精液常温保存效果最佳,有效保存时间为4d,精子活率为0.63,质膜完整率为59.33%,顶体完整率为78.29%,均显著高于其他处理组(P0.05);在6号自配基础粉中加入10μmol/L Sch B猪精液的有效保存时间为5d,精子活率达到0.61,T-AOC浓度为4.85 U/mL,显著高于其他处理组(P0.05),并使MDA浓度显著降低为1.44nmol/mL(P0.05)。因此,基础稀释粉中添加Sch B可提高猪精液常温保存品质,延长精子有效保存时间,其最佳添加量为10μmol/L。  相似文献

选用3种稀释液配方分别对6头种公牛精液进行稀释、封装、冷冻,并检查种公牛冻精解冻后的精子活力、畸形率、顶体完整率、冻后废弃率、存活时间及镜检感光度.结果表明:3种不同稀释液配方对种公牛精液冻后活力影响差异均显著(P<0.05),Ⅰ稀释液生产冻精的畸形率极显著高于Ⅱ稀释液和Ⅲ稀释液(P<0.01),3种稀释液冻精精子顶体完整率之间差异均极显著(P<0.01),Ⅲ稀释液与Ⅰ稀释液和Ⅱ稀释液间对牛精液所生产冻精的冻后废弃率影响均差异显著(P<0.05),Ⅰ稀释液与Ⅱ稀释液和Ⅲ稀释液间对牛精液所生产冻精的存活时间影响差异也显著(P<0.05),Ⅲ稀释液生产的冻精精子感光度优于Ⅰ稀释液和Ⅱ稀释液,其余各项指标差异均不显著(P>0.05).因此,可用自配Ⅱ稀释液替代进口专用Ⅲ稀释液.  相似文献

通过比较三种冷冻精液保存稀释液对冻后精子活力的影响,采用设定的工艺流程制作细管冻精.结果表明,其精子的解冻活力稀释液Ⅲ保存效果极显著优于稀释液Ⅰ的保存效果(P<0.01),稀释液Ⅱ保存效果极显著优于稀释液Ⅰ的保存效果(P<0.01),稀释液Ⅲ与稀释液Ⅱ的保存效果差异不显著(P〉0.05).总存活时间稀释液Ⅱ保存时间明显长于稀释液Ⅲ的保存时间.  相似文献

为了减小卵黄中大颗粒物质对精液品质的影响,试验使用3种稀释液在0~5℃条件下对3只蓝狐精液进行低温保存试验,每只狐精液分为4组,即Ⅰ组(原精液)、Ⅱ组(东林Ⅱ号狐常温精液稀释液)、Ⅲ组(加入20%卵黄的东林Ⅱ号狐常温精液稀释液)、Ⅳ组(加入20%卵黄的东林Ⅱ号狐常温精液稀释液经自然沉淀后的上清液)。结果表明:Ⅲ组、Ⅳ组精子存活时间、生存指数平均值与Ⅰ组相比差异极显著(P0.01),与Ⅱ组相比差异显著(P0.05),Ⅲ组精子存活时间、生存指数平均值均低于Ⅳ组但差异不显著(P0.05)。因此,加入20%卵黄的东林Ⅱ号狐常温精液稀释液经自然沉淀后的上清液低温保存蓝狐精子效果良好。  相似文献

为了筛选出效果较好的精液稀释粉,试验选择会泽县常用的三种稀释粉对大约克公猪精液按一定比例进行稀释,采用MS-FX-101A型种猪精液质量检测系统检测稀释精液的精子活力、密度,姬姆萨染色后镜检精子畸形率。结果表明:三种稀释粉对常温下种猪稀释精液的精子活力影响较大,对畸形率的影响较小;保存5 d后,Ⅰ号(日本产稀释粉)稀释精液的精子活力为(58.89±2.27)%,Ⅱ号(比利时产稀释粉)稀释精液的精子活力为(57.78±0.38)%,Ⅲ号(韩国产稀释粉)稀释精液的精子活力为(38.78±6.83)%;Ⅰ号与Ⅱ号之间差异不显著(P0.05),Ⅰ号、Ⅱ号与Ⅲ号之间差异极显著(P0.01);Ⅲ号稀释精液的精子畸形率略低,但三种稀释精液精子的畸形率差异不显著(P0.05)。说明Ⅰ、Ⅱ号稀释粉的精液保存效果较好,较适合猪人工授精站推广使用。  相似文献

为探讨课题组研制的猪精液17℃常温保存稀释剂在高倍稀释下对猪精子的保护效果,试验以BTS稀释剂为对照组,分别将原精进行1,3,5,10和15倍稀释,以精子有效存活时间内的活力、质膜完整性和顶体完整性为评价指标,研究课题组研制的稀释剂对高倍稀释的猪精液常温保存效果的影响。结果表明,配方组在5倍和10倍稀释时精液有效保存时间分别为8.77 d和8.43 d,在15倍稀释时配方组中精液有效保存时间明显下降,仅为4.56 d。而对照组中,精液最长有效保存时间为3倍稀释下的3.1 d;配方组采用10倍稀释,保存至第3天精子质膜完整率为82%,顶体完整率达87%,与对照组相比差异显著(P 0.05)。试验结论是课题组研制的稀释剂能够使猪精子耐受高倍稀释并具有较好的保护作用。  相似文献

采用高效液相色谱法测定癸氧喹酯干混悬剂的含量,在2-250μg/mL范围内,峰面积的常用对数与进样量浓度的常用对数呈良好的线性关系,R^2=1(n=5),平均回收率为99.24%~99.51%,RSD在0.05%~0.28%。此方法分析时间短,样品前处理简便、定量结果准确,重现性好,结果满意,为其质量控制提供了依据。  相似文献

以国际标准强毒R株人工感染非免疫产蛋鸡,定时扑杀,分别从鼻窦、眶下孔、气管、肺、气囊、卵巢和输卵管分离MG,并收集感染鸡所产蛋分离MG。结果表明,人工感染48小时后上、下呼吸道及肺已被全面感染,96小时气囊已被感染,120小时输卵管已能分离到MG,卵巢始终分离不到MG。人工感染鸡自144小时便能在其所产蛋中分离出MG。药物治疗能在72小时内消除感染,油乳剂苗则需24天后逐渐降低蛋内MG分离率,药物卵内注射、种蛋药浴、高温处理均能杀死卵内MG,但以研制的种蛋浸泡剂药浴效果为最好。  相似文献

REASONS FOR PERFORMING STUDY: Centesis of the bicipital bursa using an 8.9 cm long spinal needle has been reported but the alternative of employing a 3.8 cm long hypodermic needle requires validation. OBJECTIVE: To compare the efficacy of 2 different methods of centesis of the bicipital bursa and to evaluate the usefulness of ultrasonographic imaging to determine the location of solution administered when centesis of the bursa is attempted. METHODS: For Trial 1, 6 clinicians, who had no previous experience of centesis of the bicipital bursa, attempted to inject a solution composed of an aqueous radiopaque contrast medium and physiological saline solution (PSS) into the bicipital bursae of 2/12 horses using the previously described distal approach to inject one bursa and a proximal approach to inject the contralateral bursa. The bicipital tendon and bursa were examined ultrasonographically before and after injection; and both shoulders were examined radiographically to identify the location of the medium. In Trial 2, another 6 clinicians, also with no previous experience of centesis, repeated Trial 1, using 6 horses, but the radiopaque contrast medium was mixed with air instead of PSS. RESULTS: Accuracy of centesis using the proximal approach was 39% and that of the distal approach 28%. Ultrasonographic examination of the shoulder allowed the location of solution and air to be accurately predicted in all 12 shoulders examined. CONCLUSIONS: Clinicians who have had no previous experience performing centesis of the bicipital bursa are unlikely to be successful in centesis using either approach. Radiographic examination after injecting a radiopaque contrast medium may be necessary to assess the success of centesis especially if bursal fluid is not obtained during centesis. Injecting air along with the radiopaque contrast medium provides more accurate ultrasonographic confirmation of centesis and better radiographic definition than does injection without air.  相似文献

乳酸杆菌作为益生菌广泛用于人和动物。本文综述了乳酸杆菌改善宿主健康的机制。乳酸杆菌可通过产生抗菌物质如乳酸、过氧化氢、细菌素,或者通过竞争营养或肠道黏附位点来抑制致病菌;通过诱导黏附素的分泌或阻止细胞凋亡而增强肠道的屏障功能,从而保护肠道。文章重点讨论了乳酸杆菌表面成分(表面蛋白、脂磷壁酸和肽聚糖)与肠道受体(C型凝集素受体、Toll样受体和 Nod样受体),阐述了他们结合后启动免疫调节信号,调控肠道免疫功能以发挥改善健康作用的机制。  相似文献

Ingestively masticated fragments were collected and sized via sieving. Different sizes of esophageal masticate and ruminal digesta fragments, and ground fragments of larger masticated pieces were incubated in vitro, and undigested NDF remaining at intervals of up to 168 h of incubation was determined. The ruminal age-dependent time delay (tau) for onset of digestion of NDF was positively correlated (P < 0.004) with the mean sieve aperture estimated to retain 50% of the fragments between successive sieve apertures (MRA). Degradation rate of potentially degradable NDF (PDF) and level of indigestible NDF were not related (P > 0.10) to MRA of masticated and ground fragments. Estimates of tau were positively related to MRA, with slopes of bermudagrass < corn silage < ruminal fragments of corn silage. It was concluded that fragment size-, and consequently, ruminal age-dependent onset of PDF degradation of a mixture of different fragment sizes results in an age-dependent rate of degradation of the more rapidly degrading of two subentities of PDF. Models are proposed that assume a tau before onset of simultaneous degradation of PDF from two pools characterized as having gamma-modeled age-dependency and age-constant rates. The ruminal age-dependent pool seems to be associated with the faster-degrading pool, and its rate parameter increases with range in MRA in the population of fragments. Conceptually, the ruminal age-dependent rate parameter for PDF degradation seems to represent a composite of several effects: 1) effects of the size-dependent tau; 2) range in MRA of the population of ingestively masticated fragments; and 3) subentities of PDF that degrade via more rapid age-dependent rates compared with subentities of PDF that degrade via age-constant rates. The estimated fractional rates of ruminative comminution of ingestively masticated fragments (0.060 to 0.075/h) were of a magnitude similar to the mean fractional rates of PDF digestion (0.030 to 0.085/h), which implies that ruminative comminution may be first-limiting to fractional rate of PDF digestion. The in vivo roles of ingestive and ruminative mastication of fragments on PDF degradation must be considered in any kinetic system for estimating PDF digestion in the rumen. These results and others in the literature suggest that the rate of surface area exposure rather than intrinsic chemical attributes of PDF may be first-limiting to degradation rate of PDF in vivo.  相似文献
