Contagious mastitis pathogens continue to pose an economic threat to the dairy industry. An understanding of their frequency and transmission dynamics is central to evaluating the effectiveness of control programmes. The objectives of this study were twofold: (1) to estimate the annual herd-level incidence rates and apparent prevalences of Streptococcus agalactiae (S. agalactiae) in the population of Danish dairy cattle herds over a 10-year period from 2000 to 2009 inclusive and (2) to estimate the herd-level entry and exit rates (demographic parameters), the transmission parameter, β, and recovery rate for S. agalactiae infection. Data covering the specified period, on bacteriological culture of all bulk tank milk samples collected annually as part of the mandatory Danish S. agalactiae surveillance scheme, were extracted from the Danish Cattle Database and subsequently analysed. There was an increasing trend in both the incidence and prevalence of S. agalactiae over the study period. Per 100 herd-years the value of β was 54.1 (95% confidence interval [CI] 46.0-63.7); entry rate 0.3 (95% CI 0.2-0.4); infection-related exit rate 7.1 (95% CI 5.6-8.9); non-infection related exit rate 9.2 (95% CI 7.4-11.5) and recovery rate 40.0 (95% CI 36.8-43.5). This study demonstrates a need to tighten the current controls against S. agalactiae in order to lower its incidence.  相似文献

Streptococcus agalactiae mastitis persists as a significant economic problem for the dairy industry in many countries. In Denmark, the annual surveillance programme for this mastitis pathogen initially based only on bacteriological culture of bulk tank milk (BTM) samples, has recently incorporated the use of the real-time PathoProof Mastitis PCR assay with the goal of improving detection of infected herds. The objective of our study was to estimate the herd sensitivity (Se) and specificity (Sp) of both tests of BTM samples using latent class models in a Bayesian analysis while evaluating the effect of herd-level covariates on the Se and Sp of the tests. BTM samples were collected from all 4258 Danish dairy herds in 2009 and screened for the presence of S. agalactiae using both tests. The highest Se of PCR was realized at a cycle threshold (Ct) cut-off value of 40. At this cut-off, the Se of the PCR was significantly higher (95.2; 95% posterior credibility interval [PCI] [88.2; 99.8]) than that of bacteriological culture (68.0; 95% PCI [55.1; 90.0]). However, culture had higher Sp (99.7; 95% PCI [99.3; 100.0]) compared to PCR (98.8; 95% PCI [97.2; 99.9]). The accuracy of the tests was unaffected by the herd-level covariates. We propose that screenings of BTM samples for S. agalactiae be based on the PCR assay with Ct readings of <40 considered as positive. However, for higher Ct values, confirmation of PCR test positive herds by bacteriological culture is advisable especially when the between-herd prevalence of S. agalactiae is low.  相似文献

Factors associated with the presence of Mycoplasma sp. in bulk tank milk samples were evaluated from 664 herds during 2.25 years. Milk quality components were not strongly related to the presence of Mycoplasma sp. in bulk tank milk. The presence of other contagious mastitis pathogens, Staphylococcus aureus and Streptococcus agalactiae, was also not related to the presence of mycoplasma, suggesting that the aetiology and transmission of mycoplasma mastitis were different from transmission of other contagious mastitis pathogens. The occurrence of the first isolation of mycoplasma from a bulk tank was not correlated to season of the year. Mycoplasma in bulk tank milk samples were more likely to be found in herds shipping more milk, an indirect measure of herd size. This suggests that larger herds are more likely to have mycoplasma mastitis. However, the first appearance of mycoplasma mastitis in a bulk tank sample was followed by a sample without this pathogen in more than 60% of herds. Mycoplasma sp. was not detected in any herd a year after first isolation. These findings suggest that this pathogen could be controlled and eliminated from herds.  相似文献

The objective of this study was to estimate the prevalence of mycoplasma, Staphylococcus aureus, and Streptococcus agalactiae in bulk tank milk (BTM) in Québec dairy herds. BTM was sampled 3 times a month in 117 randomly selected dairy herds. Samples were submitted for S. aureus, S. agalactiae, and mycoplasma and for direct mycoplasma detection by polymerase chain reaction (PCR). Mycoplasma spp. was identified at least once in 3 herds (2.6%) by primary culture and/or PCR and in 4 herds (3.4%) by enrichment culture and/or PCR. Staphylococcus aureus was isolated at least once in 99 (84.6%) and 112 (95.7%) herds in primary culture and after enrichment, respectively. Streptococcus agalactiae was isolated at least once in 9 (7.7%) and 10 (8.6%) herds in primary culture and after enrichment, respectively. Herd prevalence of mycoplasma was similar to that previously reported in Canada. Staphylococcus aureus is still by far the most important contagious mastitis pathogen.  相似文献

Mycoplasma bovis (M. bovis) is a highly infectious pathogen of cattle causing pneumonia, polyarthritis, otitis, and less frequently, subcutaneous abscesses, abortions and meningitis. Ineffective drugs treatments, culling of infected cows and loss of milk production can lead to significant economic loss on dairy farms. The early detection of cows excreting M. bovis bacteria to prevent mastitis outbreaks is warranted. Reports suggest that the risk of M. bovis mastitis is higher in larger dairy herds. The objective of this study is to estimate the herd-level prevalence of M. bovis in Flanders, Belgium by culturing bulk tank milk samples taken from dairy farms. Three bulk tank milk samples per dairy herd were taken over four weeks, with collection intervals of two weeks. Culturing was done after pre-incubation using modified Hayflicks media to increase the chances of recovery of bacteria. For the identification of M. bovis, tDNA intergenic spacer PCR was used. In three herds (1.5%) of the 200 herds sampled, M. bovis was isolated from one of the three consecutive bulk tank milk samples. We conclude that in Flanders in 2009 at least 1.5% of the dairy herds had one or more cows excreting M. bovis in the milk. The frequent monitoring of bulk tank milk to detect the presence of M. bovis, especially in expanding herds on farms that often purchase replacement animals, should be encouraged in order to detect the presence of M. bovis and to monitor the success of control procedures following an outbreak of mycoplasmal mastitis in the herd.  相似文献

The purpose of this study was to 1) estimate the herd prevalence of contagious mastitis pathogens in bulk milk from Prince Edward Island (PEI) dairy farms, 2) determine the association between bulk milk culture results and mean bulk milk somatic cell count (BMSCC), and 3) investigate the agreement of repeated bulk milk cultures. Three consecutive bulk milk samples were obtained at weekly intervals from all 258 PEI dairy herds and were cultured using routine laboratory methods. Cumulative prevalence of Staphylococcus aureus, Streptococcus agalactiae, and Mycoplasma spp. (M. bovis and M. alkalescens) was 74%, 1.6%, and 1.9%, respectively. Bulk milk somatic cell count of Staph. aureus-positive herds was higher than that of negative herds. Agreement for Staph. aureus isolation between 3 consecutive tests was moderate (kappa = 0.46). Mycoplasma bovis and M. alkalescens in bulk milk are being reported for the 1st time in PEI ever and in Canada since 1972.  相似文献

奶牛乳房炎是引起奶牛业经济损失的一种重要疫病,目前还没有快速、特异检测奶牛乳房炎主要致病原的方法。本试验根据金黄色葡萄球菌、无乳链球菌、大肠杆菌各自保守的16S或23S rRNA基因序列,合成了3对特异性引物,建立了三重PCR检测方法。特异性试验表明,该方法对所有参与测试的金黄色葡萄球菌、无乳链球菌和大肠杆菌都能扩增出各自的阳性条带,而对所有参与测试的对照菌株则不能扩增出任何条带。敏感性试验表明该方法能检测到4个菌的金黄色葡萄球菌、无乳链球菌和2个菌的大肠杆菌。对送检的乳房炎奶样36份直接进行PCR检测,金黄色葡萄球菌阳性7份,无乳链球菌阳性2份,大肠杆菌阳性6份。  相似文献

The relation between the prevalence of contagious mastitis with the finding of Streptococcus agalactiae and Staphylococcus aureus and the number of somatic cells in the tank samples of milk was studied on the basis of a clinical and bacteriological examination of 11 129 dairy cows on four large farms, with a capacity of 600 to 1240 animals. A statistically significant correlation was demonstrated between the analyzed characteristics (r = 0.621). The relation was demonstrated at a prevalence up to 25.3%. The number of somatic cells provides a marked indication of prevalence only from 10%. We failed to demonstrate a statistically significant correlation between the prevalence of mastitis with a cultivation finding significant correlation between the prevalence of mastitis with a cultivation finding of Staphylococcus aureus and the number of somatic cells in tank samples of milk (r = 0.054). The results suggest that on large farms it will not be possible to use only the number of somatic cells in tank milk samples as a signal of diagnostic improvement and preventive mastitis-control measures, since a 10% prevalence of these diseases usually excludes the possibility of herd sanitation.  相似文献

奶牛乳房炎是奶牛常见病和多发病之一,其病因复杂,难以防制,给奶牛饲养业带来了巨大的经济损失。作者通过对某奶牛场的奶牛临床型乳房炎病原菌进行分离、鉴定,从105份样品中共分离出76株细菌,经培养特性、染色镜检、生化鉴定等试验,鉴定出无乳链球菌58株,占分离菌的76.32%;大肠杆菌18株,占分离菌的23.68%。本试验结果表明,无乳链球菌是奶牛临床型乳房炎的主要病原菌,其次是大肠杆菌。这为奶牛乳房炎的防制提供了重要依据。  相似文献

Streptococcus agalactiae continues to be a major cause of subclinical mastitis in dairy cattle and a source of economic loss for the industry. Veterinarians are often asked to provide information on herd level control and eradication of S. agalactiae mastitis. This review collects and collates relevant publications on the subject. The literature search was conducted in 1993 on the Agricola database. Articles related to S. agalactiae epidemiology, pathogen identification techniques, milk quality consequences, and control, prevention, and therapy were included. Streptococcus agalactiae is an oblique parasite of the bovine mammary gland and is susceptible to treatment with a variety of antibiotics. Despite this fact, where state or provincial census data are available, herd prevalence levels range from 11% (Alberta, 1991) to 47% (Vermont, 1985). Infection with S. agalactiae is associated with elevated somatic cell count and total bacteria count and a decrease in the quantity and quality of milk products produced. Bulk tank milk culture has, using traditional milk culture techniques, had a low sensitivity for identifying S. agalactiae at the herd level. New culture methods, using selective media and large inocula, have substantially improved the sensitivity of bulk tank culture. Efficacy of therapy on individual cows remains high. Protocols for therapy of all infected animals in a herd are generally successful in eradicating the pathogen from the herd, especially if they are followed up with good udder hygiene techniques.  相似文献

The number of somatic cells and the isolation of the causative agents of mastitis in quarter, composite, bucket, and bulk tank samples of cow's milk was determined four times during a six-month period. The number of somatic cells in milk samples indicated a degree of mastitis infection and was influenced neither by the year season nor by the length of lactation. At a repeated examination of 28 dairy cows an increased number of somatic cells in milk was found once in 68 udder quarters and with three successive samplings only in 21 quarters. The etiological agents of mastitis were detected once in 31 quarters and three times in succession only in five quarters. The number of cows positive by the number of cells in quarter samples of milk increased from 52.9-58.8% at a single examination to as much as 100% at four examinations. The etiological agents of mastitis were isolated in a single examination in 17.6% of cows and at four examinations in 58.8% of cows. The composite and bucket samples of milk containing 200 to 300 thousand cells per ml are recommended to be considered as mastitis-positive: in 68 to 78% they came from cows having more than 500 thousand cells per ml at least in one quarter sample. The number of cells in a bulk sample was in correlation with the percentage of cows having a positive NK-test (similar to CMT) and positive isolation of S. agalactiae from quarter milk samples.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Samples of bulk tank milk and cow-composite milk from 23,138 dairy cows from 50 California dairies were examined by use of microbiologic procedures. The number of colonies of mastitis pathogens isolated per milliliter of bulk tank milk (used as a predictor of the percentage of infected cows in the herd) was evaluated, using simple regression analysis and Spearman's rank correlation. Correlations between the pathogens and the percentage of cows in each herd shedding the pathogens were found for Streptococcus agalactiae (r = 0.71) and mycoplasma (r = 0.59), but were considerably lower for other pathogens. When greater than or equal to 4,000 colonies of Streptococcus agalactiae were found per milliliter of bulk tank milk, at least 7% of the cows in the herd was shedding this organism. However, a pattern was not found between the number of mycoplasma colonies per milliliter of bulk tank milk and the percentage of infected cows in the herd.  相似文献

An assay was developed and evaluated for screening for Staphylococcus aureus in milk samples from cases of bovine mastitis by overnight cultivation in a broth containing 7.5 per cent sodium chloride, followed by pcr to amplify the nuc gene. The assay could detect concentrations of S aureus as low as 1 colony-forming unit/ml milk. Among 106 milk samples collected from individual quarters of lactating cows in one dairy herd and from a bulk tank, S aureus was detected in nine samples by the pcr assay but in only three samples by conventional microbiological culture.  相似文献

In a cross-sectional study of 400 randomly selected smallholder dairy farms in the Tanga and Iringa regions of Tanzania, 14.2% (95% confidence interval (CI)=11.6-17.3) of cows had developed clinical mastitis during the previous year. The point prevalence of subclinical mastitis, defined as a quarter positive by the California Mastitis Test (CMT) or by bacteriological culture, was 46.2% (95% CI=43.6-48.8) and 24.3% (95% CI=22.2-26.6), respectively. In a longitudinal disease study in Iringa, the incidence of clinical mastitis was 31.7 cases per 100 cow-years. A randomised intervention trial indicated that intramammary antibiotics significantly reduced the proportion of bacteriologically positive quarters in the short-term (14 days post-infusion) but teat dipping had no detectable effect on bacteriological infection and CMT positive quarters. Other risk and protective factors were identified from both the cross-sectional and longitudinal included animals with Boran breeding (odds ratio (OR)=3.40, 95% CI=1.00-11.57, P<0.05 for clinical mastitis, and OR=3.51, 95% CI=1.29-9.55, P<0.01 for a CMT positive quarter), while the practice of residual calf suckling was protective for a bacteriologically positive quarter (OR=0.63, 95% CI=0.48-0.81, P相似文献

A serological survey for Neospora caninum and Besnoitia besnoiti was carried out in beef and dairy cattle in South Australia. Serum samples of dairy cattle (n=133) from 9 properties and tank milk samples from a further 122 dairy herds were tested. An additional 810 sera from beef cattle from 51 properties were also tested. Testing at the individual animal level by IDEXX NEOSPORA X2 Ab test ELISA revealed a low prevalence of N. caninum antibodies of only 2.7% (95% CI; 1.6-3.7%) sera positive, as did the milk testing that showed 2.5% (95% CI; 1.4-3.6%) of tank milks being positive. At the herd level, 29.4% (95% CI; 16.9-41.9%) of beef, and 44.4% (95% CI; 12.0-76.9%) of dairy cattle herds showed serum antibodies. The highest within-herd prevalence in beef was 20% and 25%in dairy, which explains the low herd prevalence in dairy detected by bulk milk testing. Testing for B. besnoiti antibodies by PrioCHECK(?) Besnoitia Ab 2.0 ELISA initially identified 18.4% (95% CI: 15.8-21.0%) of 869 individual cattle sera as positive by ELISA at the manufacturer's suggested cut-off threshold (15 PP). Additional tests by immunoblot and IFAT, however, could not confirm any of the ELISA results. The use of a higher (40 PP) threshold in the ELISA is suggested to improve specificity. There is thus no evidence of B. besnoiti infection in South Australian cattle.  相似文献

无乳链球菌是导致奶牛乳房炎的常见病原菌之一,为了掌握青海地区无乳链球菌的流行情况及常用药物的敏感性,从而为本地区更好的预防与治疗奶牛乳房炎提供保障。本试验在青海地区16个奶牛场共采集258份乳房炎乳样,经革兰氏染色与绵羊鲜血琼脂培养基培养纯化,获得疑似链球菌137株;随后经玻片法血浆凝固酶试验、CAMP试验、生化试验与动物致病性试验对无乳链球菌进行鉴定,结果显示,137株链球菌中含有无乳链球菌42株,分离率为30.7%;经K-B纸片扩散法对无乳链球菌耐药性检测结果显示,该地区无乳链球菌主要对头孢类高度敏感,对氧氟沙星、磺胺类、氨苄青霉素与阿米卡星中度敏感。  相似文献

Thirty-two dairy herds, 16 with low somatic cell counts (LSCC; Dairy Herd Improvement Association 12-month mean herd SCC less than or equal to 150,000 cells/ml) and 16 with high somatic cell counts (HSCC; Dairy Herd Improvement Association 12-month mean herd SCC greater than or equal to 700,000 cells/ml) were evaluated to determine the relationship between the prevalence of mastitis in each herd and each herd's mastitis control and management practices. Once for each herd, duplicate quarter milk samples were collected from the lactating cows, a survey of herd mastitis control, milking hygiene, and management practices of each herd was performed, and milking-machine function was evaluated. Of the 16 herds with LSCC, 2 (12.5%) had Streptococcus agalactiae isolated and 7 (44%) had Staphylococcus aureus isolated. Both organisms were found in all of the herds with HSCC. In herds with LSCC, the mean percentage of quarters infected with Str agalactiae was 0.1%, the mean percentage infected with streptococci other than Str agalactiae was 1.9%, and the mean infected with S aureus was 0.7%. In herds with HSCC, 25.7% of the quarters were infected with Str agalactiae, 3.7% were infected with streptococci other than Str agalactiae, and 7.6% were infected with S aureus. A program of postmilking teat dipping and treatment of all cows at the beginning of the nonlactating period was practiced more frequently in the herds with LSCC (81.3%) than in the herds with HSCC (37.5%). Major differences were not found between the 2 groups of herds in the use of the more common milking hygiene techniques or in the maintenance and functional characteristics of the milking equipment.  相似文献

用兰州乳房炎检测试剂(Lanzhou mastitis test,LMT)对克拉玛依规模化奶牛场进行了奶牛隐性乳房炎流行病学调查。结果显示,该地区奶牛隐性乳房炎的发病率为70.43%(543/771)。对543份阳性牛乳样品进行了病原菌的分离鉴定,结果发现,阳性乳样品中细菌分离率达89.13%(484/543),葡萄球菌、链球菌、肠杆菌和棒状杆菌为主要致病菌,且多为几种致病菌的混合感染。  相似文献

The objective of this study was to describe the frequency of occurrence of clinical mastitis in dairy herds in Ontario. The study group consisted of 65 dairy farms involved in a 2-year observational study, which included recording all clinical mastitis cases and milk sampling of quarters with clinical mastitis. Lactational incidence risks of 9.8% for abnormal milk only, 8.2% for abnormal milk with a hard or swollen udder, and 4.4% for abnormal milk plus systemic signs of illness related to mastitis were calculated for 2840 cows and heifers. Overall, 19.8% of cows experienced one or more cases of clinical mastitis during location. Teat injuries occurred in 2.1% of lactations. Standard bacteriology was performed on pretreatment milk samples from 834 cows with clinical mastitis. The bacteria isolated were Staphylococcus aureus (6.7%), Streptococcus agalactiae (0.7%), other Streptococcus spp. (14.1%), coliforms (17.2%), gram-positive bacilli (5.5%), Corynebacterium bovis (1.7%), and other Staphylococcus spp. (28.7%). There was no growth in 17.7% of samples, and 8.3% of samples were contaminated. Clinical mastitis is a common disease in dairy cows in Ontario; approximately 1 in 5 cow lactations have at lease one episode of clinical mastitis. There is, however, considerable variation in the incidence of clinical mastitis among farms. The majority of 1st cases of clinical mastitis occur early in lactation, and the risk of clinical mastitis increases with increasing parity. Environmental, contagious, and minor pathogens were all associated with cases of clinical mastitis.  相似文献
