The atomic structure of dislocation cores is decisive for the understanding of plasticity in crystalline solids. The core structure of dislocations in sapphire introduced by high-temperature plastic deformation has been investigated with the use of the negative spherical-aberration imaging technique. The ability of this technique to discriminate oxygen columns from aluminum (Al) columns, combined with reproduction of subtle contrast features by image simulation, leads to a markedly detailed atomic model of the dislocation cores. The partial dislocations are Al-terminated, with electrical neutrality being achieved because half of the Al columns are missing. These partials also undergo core spreading, which results in random occupancy of both tetrahedrally and octahedrally coordinated sites, though Al in tetrahedral coordination never occurs in a perfect crystal. Unusual dislocation core structures may be present in other technologically important nonmetallic solids.  相似文献

This study aimed to develop a mathematical model describing the dynamics of paratuberculosis (PTB) in red deer (Cervus elaphus) under pastoral farming conditions in New Zealand. The model examined infectivity differences between ovine and bovine strains of Mycobacterium avium subspecies paratuberculosis (MAP) and seasonality of MAP survival. We also evaluate variable use of pasture and the effect of management interventions on the infection prevalence and annual clinical incidence of PTB. A state-transition model was developed and calibrated to observed data on both prevalence of infection and incidence of clinical PTB. To accommodate specific PTB features for deer, the model included a fast and a slow track for progression of infection to disease. MAP on pasture was the source for horizontal transmission and infected dams for vertical transmission. In the presence of a single strain, an infectivity reduction of up to 80% allowed MAP to persist in the herd (R(0)>1). For mixed infection by two strains however, a 30% reduction in infectivity of one strain was sufficient to outcompete a strain with lower infectivity, suggesting that mixed infection of MAP strains with different infectivity may not be common in deer. The model showed that seasonal variation of MAP survival on pasture had little impact on transmission dynamics, and that rotational grazing with pasture spelling vs. permanent grazing of the same paddock reduced both infection prevalence and clinical PTB by about 50%. Based on model outputs, early detection of young deer in a high-shedding state was the most effective means of controlling PTB among the tested scenarios.  相似文献

Large amounts of veterinary antibiotics enter soil via manure of treated animals. The effects on soil microbial community structure are not well investigated. In particular, the impact of antibiotics in the presence of manure is poorly understood. In this study, two agricultural soils, a sandy Cambisol (KS) and a loamy Luvisol (ML), were spiked with manure and sulfadiazine (SDZ; 0, 10 and 100 μg g?1) and incubated for 1, 4, 32 and 61 days. Untreated controls received only water. The microbial community structure was characterised by investigating phospholipid fatty acids (PLFA) and using PCR–denaturing gradient gel electrophoresis (DGGE) of 16S rDNA. The total concentration of PLFA increased with addition of manure and was reduced by both SDZ concentrations at incubation times >4 days. The SDZ addition decreased the bacteria:fungi ratio. The largest stress level, measured as ratio of PLFA (cyc17:0 + cyc19:0)/(16:1ω7c + 18:1ω7c), was found for the controls, followed by the manure treatments and the SDZ treatments. A discriminant analysis of the PLFA clearly separated treatments and incubation times. Both soils differed in total PLFA concentrations and Gram?:Gram+ ratios, but showed similar changes in PLFA pattern upon soil treatment. Effects of manure and SDZ on the bacterial community structure were also revealed by DGGE analysis. Effects on pseudomonads and β-proteobacteria were less pronounced. While community structure remained altered even after two months, the extractable concentrations of SDZ decreased exponentially and the remaining solution concentrations after 32 days were ≤27% of the spiking concentration. Our results demonstrate that a single addition of SDZ has prolonged effects on the microbial community structure in soils.  相似文献

The aim of this study was to determine potential cumulative effects of repeated passes with current heavy agricultural machinery on topsoil (0–0.3 m) and subsoil (below 0.3 m) physical properties of a Luvisol as affected by long-term tillage (annual mouldboard ploughing to 0.3 m depth (MP), shallow-mixing conservation tillage to 0.1 m depth (SM) with a wing-bladed rigid tine cultivator). Moreover, sugar beet yield was determined. Wheeling was conducted with a six-row self-propelled sugar beet harvester representing contemporary heavy agricultural machinery (wheel load 7.8–11.7 Mg, average ground contact pressure 100–145 kPa). Wheeling was applied once per year over three consecutive years after harvest of sugar beet, cereal and cereal, and moreover, independent from regular plot management with light experimental machinery. Soil moisture at wheeling (0–0.6 m depth) was around 100% field capacity in most years, which was secured by irrigation before wheeling if necessary.Repeated wheeling negatively affected penetration resistance, macropore volume (equivalent diameter >50 μm) and air permeability of topsoil (0.05–0.1 m, 0.18–0.23 m) and subsoil (0.4–0.45 m) layers, while biopore number and surface water infiltration remained unaffected. SM compared to MP tillage increased penetration resistance while decreasing macropore volume and air permeability in the 0.18–0.23 m layer, whereas reverse effects occurred in 0.4–0.45 m depth. Sugar beet yield was decreased by wheeling and SM tillage compared to the control treatments. No significant interactions between wheeling and tillage occurred in any parameter investigated.Conclusively, SM tillage did not provide better subsoil resistance against compaction compared to MP treatment under wheeling and soil conditions prevalent in our experiment. Repeated wheeling with heavy agricultural harvest machinery is obviously at risk to exceed the bearing capacity of susceptible soils. Although (i) under regular harvest conditions just small parts of arable fields (except headlands) are wheeled with high loads, (ii) harvest is by far not every year conducted under high soil moisture, and (iii) effects in the subsoil were small, such risks have to be taken into account. Reduction of tillage depth to <0.1 m is not recommended for high yielding sugar beet crops grown on loessial soils.  相似文献

AIM: To report current farmer opinions and farming practices relating to control of gastrointestinal nematodes and anthelmintic resistance on sheep farms in New Zealand. METHODS: An interview-based cross-sectional study of grazing management and anthelmintic usage was conducted by veterinarians on 80 randomly selected sheep farms in New Zealand. RESULTS: Useable data were returned by 74/80 (92%) farmers who participated in the study. However, despite contacting 400 farmers the target sample size of 100 farms was not reached. The results indicated that only 31% of farms had previously tested for drench resistance, that effective quarantine-drenching of imported stock was not always carried out, and that farmers were more likely to integrate cattle than ewes into their grazing management of lambs. Furthermore, the number of drenches given to lambs had changed little in 25 years. The use of faecal egg counting by farmers has increased. CONCLUSIONS: Dependence on anthelmintics continues to be high on sheep farms in New Zealand. Whilst the number of drench treatments has changed little, there is more widespread use of persistent or long-acting treatments. Farmers need to be encouraged to monitor the resistance status of nematode populations on their farms and use this information to develop strategies aimed at maintaining susceptible alleles within the parasite populations and conserving the efficacy of existing drug families.  相似文献

AIM: To determine the prevalence of the two most commonly diagnosed pathogenic Leptospira spp. serovars, Hardjobovis and Pomona, in sheep in a sheep-only abattoir in New Zealand, and to determine the prevalence of kidneys which were leptospire culture-positive collected from sheep seropositive or seronegative to the microscopic agglutination test (MAT). METHODS: A repeated cross-sectional observational study was conducted of serological and kidney culture prevalences of Leptospira borgpetersenii serovar Hardjobovis and Leptospira interrogans serovar Pomona. Lines of sheep and individual sheep were systematically randomly selected at a sheep-only abattoir during 18 May 2004 to November 2004 and 06 December 2004 to 14 June 2005. Additionally, a cross-sectional study examined prevalences in a purposively selected line of sheep from a flock with clinical evidence of an outbreak of leptospirosis. RESULTS: In the study population of 15,855 sheep of which 2,758 were sampled, 5.7 (95% CI=4.9-6.7)% were seropositive to one or both serovars; 44.2 (95% CI=34.6-54.2)% of 95 lines of sheep and 44.9 (95% CI=35.0-55.3)% of 89 farms showed serological evidence of infection. The serological prevalence of serovar Hardjobovis was significantly higher than that of serovar Pomona both at line (33% and 4%, respectively) and individual (5% and 1%, respectively) levels. A low but persistent seroprevalence of Hardjobovis throughout both years suggested low-level endemicity to this serovar, whereas Pomona infections appeared to be sporadic. Leptospires were isolated from kidneys of 8/37 (22%) Hardjobovis- and 1/6 (17%) Pomona-seropositive, and 5/499 (1%) seronegative animals. Of the animals purposively sampled from a farm with a clinical outbreak of leptospirosis, all kidneys from the 13 seropositive animals were culture-positive, indicating a high risk of exposure of meat workers in outbreak situations. Kidneys of MAT-seropositive sheep were 21.7 (95% CI=7.6-61.9) times more likely to test culture-positive than kidneys from animals with negative MAT titres. In general, the results indicated that 13/1,000 sheep slaughtered were potentially shedding leptospires. CONCLUSIONS: The study demonstrated the presence of a definite risk of occupational exposure of meat workers in a sheep-only slaughterhouse to the two most commonly diagnosed pathogenic Leptospira spp. serovars in New Zealand.  相似文献

Electron-transparent sections of the Allende meteorite, a carbonaceous chondrite, have been prepared by ion-thinning and examined by high-voltage (800-kilovolt) transmission electron microscopy. The matrix crystals, mainly olivine, range in size from approximately 5 to approximately 0.01 micrometers; carbon is present as intergranular films of poorly crystalline graphite. The chondrules exhibit extensive radiation damage, a feature lacking in the matrix. In addition, both chondrules and matrix are undeformed and contain negative crystals; submicroscopic exsolution lamellae are present in pyroxenes. Comparison of the substructure in the Allende meteorite with that in the Parnallee meteorite and in lunar and selected terrestrial rocks leads to the conclusion that chondrule irradiation preceded cold accretion during formation of the solar system and that the meteorite has since been undisturbed.  相似文献

The internal substructures of a type B sample have been examined at high magnification and compared with terrestrial rocks. Selected ultrathin sections were prepared from these multiphase materials by an ion-thinning technique and examined in a 1-Mev electron microscope, with complementary optical analyses. The structures in the ilmenite and plagioclase indicate that the lunar material has undergone plastic deformation by dislocation movement and possibly microtwinning, with subsequent recovery. The pyroxene exhibits complex lamellar structures of submicron spacing. These various observations are consistent with the optical microscopy evidence for distortion and recovery and identify the processes involved.  相似文献
