致病性猪源肠球菌16S rDNA-RFLP研究及系统进化分析 总被引:1,自引:0,他引:1
为探讨分离自河南滑县、武陟等12个地区的猪源肠球菌的亲缘关系,采用对其16SrDNA基因的RFLP分析、序列分析及系统进化树的构建等方法,对12株疑似猪源肠球菌进行了研究;结果显示,12株分离株均与肠球菌同源性最高,同源率达98.1%~100%;而与猪链球菌同源率最高达85.5%,且进化树中12株菌株均位居与猪链球菌并列的肠球菌主干分支中。HandⅢ、XbaⅠ、EcoRⅠ分别对12株球菌的16SrDNA-RFLP分析发现,它们在该片段上有限制性位点,但没有产生限制性片段长度多态性;而HinfⅠ单酶切结果出现了限制性片段长度多态性,且12株分离株可分为两大rDNA图谱类群,与实际测序分析的酶切位点相一致,且找到鉴定肠球菌的350bp的分子标记。说明所分离12株球菌均是肠球菌,且不同地理种群已出现分化。 相似文献

[目的]旨在通过黑毛和牛(Japanese Blackcattle)育肥后期投服乳酸菌制剂Entero—coccus faecalis FK-23粉末,探讨付与牛肉的附加价值和该制剂的实用性。[方法]供试牛10头,随机分为试验组和对照组,试验组5头,每头每日投服乳酸菌制剂8.0g。测量体重(BW),静脉采血检测白细胞数(WBC),维生素A(VA),维生素E(VE).总胆固醇(TC),谷草转氨酶(GOT),血尿素氮(BUN)及血糖(GLC)浓度;HPLC测验中性脂肪的脂肪酸成分。[结果]试验组与对照组比较。食欲亢进,日增重(DG)明显增加(P〈0.05),血清VE和TC明显升高(P〈0.05),VA维持预计水平走向不变。中性脂肪的不饱和脂肪酸比例出现增高趋势,出栏体重增长4.3%,胴体重增加9.3kg。[结论]结果提示,黑毛和牛育肥后期投服乳酸菌E.faecalisFK-23制剂可以获得VE含量较高的高附加价值牛肉并能提高其生产性能。 相似文献

Fifty-four quinupristin/dalfopristin-resistant Enterococcus faecium (QDREF) isolated from chickens and pigs during 2002-2003 in Korea were screened by PCR for the presence of streptogramin resistance genes vatD, vatE, and vgbA, and macrolide resistance gene ermB. None of the QDREF isolates carried vgbA and vatD genes, while vatE and ermB were detected in 9.2% and 74% of the isolates, respectively. Twenty-six percent (14/54) of the QDREF isolates contained none of the resistance determinants tested. Pulsed-field gel electrophoresis (PFGE) patterns revealed high heterogeneity: 47 different patterns for 54 QDREF evaluated. Identical PFGE types were observed in two pairs of chicken isolates and a pair of pig isolates, respectively, but chicken isolates did not share PFGE pattern with pig isolates, suggesting clonal spread of QDREF strain between the same species of animals but not between different species of animals. This is the first report, to our knowledge, of vatE-positive E. faecium isolates and also the first evidence of clonal spread of QDREF strain between animals in Korea. 相似文献

为确定导致通辽某鹅场雏鹅败血症的病原,本研究将从病鹅心、肝、淋巴结和脑组织液中分离的革兰氏阳性球菌纯化培养,进行形态学和生长耐受性观察,菌属区别、分群和菌种鉴定试验,16S rDNA检测、系统进化分析和致病性试验。结果表明,分离菌株(HQ831431)为中等致病力粪肠球菌,具有β型溶血特性,可致小鼠急性败血症。分离菌株与参考菌株ATCC29212及国内外分离的其他15株粪肠球菌16S rDNA的同源性均在99.0%以上,位于系统发育树的同一分支。 相似文献

Enterococcus faecium strains of clonal complex (CC) 17 were isolated from domestic dogs. The strains were more prevalent in infectious isolates than in colonized isolates, suggesting that strains of the CC17 lineage may have an advantage in causing infections in dogs. The pulsed field gel electrophoresis patterns of some dog and human isolates were over 90% similar. However, antimicrobial resistance patterns and virulence factors were not identical, which might reflect different use of antimicrobials in veterinary medicine or in host specificity. 相似文献

为获得产细菌素的乳酸菌,本研究对自然腌制臭鸡蛋中的乳酸菌进行分离筛选;并进行抑菌、有机酸排除、过氧化氢排除和蛋白酶敏感性试验,及温度、pH、表面活性剂和金属离子对抑菌物质的影响研究。从样品中共获得27株乳酸菌,产细菌素的乳酸菌4株,其中菌株FJY-1抑菌能力最强。经形态学、生理生化与16S rDNA基因序列分析,鉴定该菌株为屎肠球菌。其无细胞上清液在pH 2~9范围内处理2h后能维持60%以上的抑菌活性,对热稳定(121℃,30min),经胃蛋白酶处理后抑菌活性消失;该细菌素对沙门氏菌、铜绿假单胞菌、金黄色葡萄球菌等均有较强的抑制作用,并证明其对指示菌的作用方式是杀菌,该细菌素在食品防腐中具有较强的应用潜力。 相似文献

从神农架金丝猴的粪便中分离出1株益生菌株dlt7a,经鉴定为粪肠球菌。为进一步提高发酵液中dlt7a的活菌数量,对菌株dlt7a的发酵工艺进行优化。通过单因素水平试验,确定dlt7a的最佳培养条件为装液量20%(50mL/250mL),接种量2%,培养基初始pH 6.5。设计Plackett-Burman(PB)试验和响应面试验,找出发酵培养基中影响dlt7a活菌数的主要成分是蔗糖、鱼粉和KH_2PO_4,并优化3种重要成分的含量,确定最优发酵培养基配方为:蔗糖2.55%,鱼粉2.63%,豆粕2.5%,酵母膏0.4%,CaCO_3 1.0%,KCl 0.1%,MgSO_4·7H_2O 0.1%,KH_2PO_4 0.014%。在此条件下培养dlt7a菌株,活菌数可达58.83×108cfu/mL,比优化前提高了46.53%。 相似文献

17 feces samples of yak which were collected in Hongyuan county were measured with Gram staining method and 16S rRNA molecular identification in this study.8 suspected Enterococcus were separate from feces samples by bacteria purification and PCR amplification with 1 500 bp specific band. 6Enterococcus faecalis and 2Enterococcus faecium were identified through 16S rRNA sequencing.The homology analysis of the strains revealed that the homology between Enterococcus faecalis and reference strains sequence were 99.7% to 100%,that of Enterococcus faecium and reference sequence were 98.2% to 99.2%,indicating that the yak Enterococcus was highly conserved in the process of genetic evolution.The drug sensitive test results showed that the isolated strains were highly resistance to aminoglycoside antibiotics.Enterococcus faecium 11-1-2 strain was not only 5 multi-resistant,but also showed resistence to vancomycin.Enterococcus faecalis strains was most 3 multi-resistant.The antibiotics resistance results revealed that the resistance of yak Enterococcus was serious and should be taken seriously. 相似文献
