猪白细胞干扰素效价测定方法的探讨   总被引:1,自引:0,他引:1  
1989年杨云楼等、1988年吴淑华等用人羊膜细胞(WISH)、猪肾原代细胞、猪肾传代细胞对比测定了猪干扰素及人干扰素的抗病毒活性,结果很接近,认为猪与人干扰素之间有较大同源性,并与1979年Carter的报道一致。参照前人的方法,我们反复探讨、摸索出猪白细胞干扰素效价测定方法,现报道于后。 1 攻击病毒(VSV:水泡性口炎病毒)的制备 1.1 选9~10日龄无鸡新城疫抗体的健康鸡胚,去头、内脏,用Hank's液洗一次,剪碎,加0.1%胰酶,37℃水浴消化20分钟,弃去胰酶液,用Hank's液再洗一次,加入生长液(含5%小牛血清的D—MEM  相似文献

干扰素(Interferon,IFN)是一种糖蛋白。由致敏猪淋巴细胞产生的商品干扰素制剂称为猪白细胞干扰素,是一种高活性、多功能的生物活性制品。干扰素是抗病毒感染的非特异性免疫的一个重要因素,能干扰几乎所有病毒的复制,保护未感染细胞抵抗病毒的攻击。干扰素对于免疫应答也有显著的调节作用,并可抗肿瘤。干扰素虽然是一种广谱的抗病毒活性物质,但却具有较严格的种属特异性。为阐明猪白细胞干扰素对其他种属动物的抗病毒作用,我们从1998-10~1999-01期间,选用四川世红生物技术有限公司生产的猪白细胞干扰…  相似文献

对猪白细胞干扰素在细胞培养中干扰牛病毒性腹泻/粘膜病BVD/MD)病毒的作用进行了试验。猪白细胞干扰素在500u/ml以上浓度可完全抑制牛病毒性腹泻/粘膜病(BVD/MD)病毒所产生的细胞病变,在25u/ml浓度可部分抑制病毒所产生的细胞病变,在250u/ml以下浓度不能抑制病毒所产生的细胞病变,但可使细胞产生细胞病变的时间延长,病变程度减轻,本试验证实干扰素在猪和牛之间存在交叉活性。  相似文献

为了观察纳米蜂胶黄酮对猪细小病毒(PPV)灭活疫苗免疫应答的影响,试验以家兔为模型动物,并以油乳剂为对照,将30只家兔随机均分为3组,第1~2组分别背部皮下注射0.5 m L纳米蜂胶黄酮和油乳剂猪细小病毒灭活疫苗,第3组为对照组注射0.5 m L生理盐水,检测血清抗体效价并观察对体液免疫的影响,测定淋巴细胞增殖及白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-6(IL-6)和γ干扰素(IFN-γ)含量并评价对细胞免疫的影响。结果表明:纳米蜂胶黄酮、油乳剂均能提高家兔对PPV灭活疫苗的免疫应答能力,油乳剂提高抗体的效果好于纳米蜂胶黄酮,纳米蜂胶黄酮促进T淋巴细胞增殖和提高细胞因子含量的效果优于油乳剂。说明纳米蜂胶黄酮能增强猪细小病毒灭活疫苗的免疫效果,是一种效果良好的疫苗佐剂。  相似文献

选择30日龄健康荣昌仔猪12头,每组3头分为4组。选择盐酸左旋咪唑、猪基因工程干扰素及蜂胶液作为免疫接种猪细小病毒疫苗的免疫增强剂,按相同的免疫程序免疫接种试验猪1个月,于免疫接种前采集血液后,以后每隔7d采集一次血液制备血清,检测猪细小病毒抗体效价。比较各组猪细小病毒抗体水平消长曲线情况,分析3种免疫增强剂的免疫增强效果。结果表明,蜂胶作为猪细小病毒疫苗的免疫增强剂效果最好,平均滴度达1∶256;左旋咪唑次之,平均滴度为1∶128;干扰素较差,滴度介于1∶64~1∶128。统计分析表明,试验组及单一细小病毒疫苗对照组免疫接种前后猪细小病毒抗体效价差异极显著(P<0.01),3种免疫增强剂与单一细小病毒疫苗对照组差异显著(P<0.05),其中,蜂胶与单一细小病毒疫苗对照组及与干扰素组差异极显著(P<0.01),盐酸左旋咪唑与干扰素组及与蜂胶组间的抗体效价差异均不显著(P>0.05),干扰素与蜂胶组的猪细小病毒抗体效价差异显著(P<0.05)。  相似文献

γ-干扰素(interferon gamma,IFN-γ)是干扰素的一种,又称为Ⅱ型干扰素或免疫干扰素,主要由活化的T细胞和NK细胞产生。IFN-γ是动物机体最重要的细胞因子之一,是主要的巨噬细胞活化因子(macrophage activating factor,MAF),具有强大的免疫调节作用和抗病毒、抗肿瘤作用。目前,干扰素基因工程和基因治疗研究引起了广泛关注,已被应用于人类疾病临床治疗和动物疾病防控研究。作者就IFN-γ的基本特征、生物学活性、检测及其在猪繁殖与呼吸综合征(PRRS)中的研究应用进行了综述。  相似文献

干扰素是一类仅由脊椎动物单核细胞和淋巴细胞在受到病毒或其他诱生剂的作用时产生的一种具有抗病毒、抗肿瘤、免疫调节等多种生物学活性的分泌性可溶糖蛋白。随着基因工程方法的发展,外源表达干扰素已经进入了工业化生产阶段,并且大量投放市场,在医药、生物和农业等多个行业领域中广泛应用。近年来,随着人们对禽类干扰素研究的逐步深入,干扰素在一些禽类病毒性疾病及肿瘤性疾病治疗方面取得的良好进展。本文现就家禽干扰素的分类,家禽干扰素的抗病毒活性以及家禽基因工程重组干扰素的应用做以综述。  相似文献

The ability of nine strains of avian infectious bronchitis virus (IBV) to induce chicken interferon has been investigated using Semliki Forest virus for the tests. The Beaudette, H120 and Connecticut 46 strains induced interferon in the allantoic fluids of embryonated hens' eggs, the highest titre (1 : 30) being associated with Beaudette; but these as well as the Massachusetts M-41 and H52 strains failed to yield interferon in primary monolayer cultures of chick kidney cells as did all nine strains in organ cultures of chick embryo trachea. None of six strains of IBV investigated was susceptible to the inhibitory effects of chicken interferon.  相似文献

Genetic variation at intron 1 of the interferon gamma gene has recently been associated with variation in faecal egg count in Romney, Merino and Soay breeds of sheep. The Texel breed is more resistant to gastrointestinal nematode infection than the Suffolk breed, based on faecal egg count. Hence, the objective of this experiment was to characterise the polymorphisms of intron 1 of the interferon gammagene in Suffolk and Texel sheep, and to determine if the characterised haplotype variants were associated with faecal egg count variation in these breeds. Intron 1 haplotypes and faecal egg measurements were determined for Suffolks (n=113) and Texels (n=135). Four haplotypes were identified in Suffolks (A, B, C and D), two haplotypes as previously described, and two previously unknown haplotypes. However, there was no association between interferon gamma intron 1 haplotype and faecal egg count in Suffolks. In contrast, two previously identified haplotypes (A and B) were present in the Texel breed, and the B haplotype was associated with resistance to nematode infection (P=0.02). These results suggest that intron 1 of the interferon gamma gene has an important role in resistance to nematode infection in the Texel breed.  相似文献

The interferon system is part of the innate immune system in vertebrates. It represents the first line of host defence against viral infections. Virus entry triggers intracellular signalling pathways which lead to the secretion of soluble factors such as interferons and other cytokines. Interferons signal to neighbouring cells that a viral infection has occurred and induce an "antiviral state" resulting in inhibition of virus replication. The first recombinant interferons were produced in the 1980ies and were considered to be a major breakthrough. At present, interferons are routinely used in the therapy of certain viral and autoimmune diseases as well as for neoplastic disorders in man. In 2001 the first interferon preparation for veterinary use was licensed in the European Union. This review summarises the molecular mechanisms of the interferon system and the viral counteractions. The current type I interferon therapies in humans are described and an overview of recent clinical studies in veterinary medicine, including cat, dog, horse, cow, sheep, pig, and poultry, is given. We review the potential application of interferons and arguments in favor or against its therapeutic use in veterinary medicine.  相似文献
