在改良的基础培养液中添加HA,主要探讨HA对牛卵母细胞成熟的影响。结果表明,当mTCM-199中HA浓度为3.0 mg/mL时,第一极体排出率和卵裂率显著高于其他各组 (P<0.05),初步表明3.0 mg/mL是无血清培养中HA的最佳浓度。当在mTC-199中添加 HA、BSA和OCS时,OCS组的第一极体排出率显著高于其他两组(P<0.05),分别为 83.40%、84.33%和92.17%,但HA组与BSA差异不显著(P>0.05);卵裂率三者差异均不显著(P>0.05);表明在无血清培养中HA可代替BSA,但是培养的效果不如血清。HA在 mTCM-199和mSOF基础培养基中,对卵母细胞的第一极体排出率和卵裂率无明显差异(P >0.05)。表明在无血清培养下,HA在复杂和简单的培养液中都支持卵母细胞的成熟。 相似文献

用切割法采集卵泡液,收集卵丘一卵母细胞复合体(Cumulus oocytes comlexs,COCs)和自然裸卵,将部分COCs去除卵丘细胞获得机械裸卵,COCs放入体外成熟培养液中培养为成熟卵母细胞,加入获能的精子液,进行体外受精。结果表明:卵母细胞的体外成熟率和卵裂率与卵泡直径密切相关,大卵泡(80.95%,P〈0.01)和中等卵泡(75.50%,P〈0.05)的卵母细胞成熟率高于小卵泡(50.27%);犬卯泡(53.53%)和中等卵泡(47.13%)的卵裂率显著高于小卵泡的32.26%(P〈0.05)。COCs、机械裸卵和自然裸卵的体外成熟率分别为75.0%、54.2%和10.5%,差异极显著(P〈0.01),卵裂率分别为53.8%、10.8%和0%,差异极显著(P〈0.01)。对照组和1×10^5、1×10^6个/mL颗粒细胞组卵母细胞体外成熟率分别为68.6%、69.6%和67.8%,无显著差异(P〉0.05),但均显著高于1×10^7个/mL(51.5%,P〈0.05)和1×10^10个/mL(35.5%,P〈0.05)颗粒细胞组,但各组间的体外受精率无显著差异(P〉0.05)。结果提示,大卵泡和中卵泡的卵母细胞的体外成熟率和卵裂率显著高于小卵泡,体外成熟培养液中添加高浓度的颗粒细胞能显著抑制卵母细胞的体外成熟。 相似文献

文章主要从采集方法、性周期阶段以及培养液中FBS浓度3个方面对牛卵母细胞体外成熟的影响进行了分析。结果表明:①不同采卵方法的采卵效果存在差异,但成熟培养和体外受精情况差异不显著(P〉0.05);②从卵泡期和黄体期卵巢采集到的卵母细胞的体外成熟率和卵裂率分别,为59.7%,56.6%和39.5%、36.6%,没有显著差异(P〉0.05);③培养液中添加20%FBS组的卵母细胞成熟率和卵裂率显著高于10%FBS和30%FBS组(P〈0.05). 相似文献

试验旨在探讨藏红花素(crocin)的抗氧化应激作用对小鼠卵母细胞体外成熟及后续胚胎发育能力的影响。在体外成熟(IVM)培养液中添加不同浓度藏红花素(0、5、10、15、20、25、30 μmol/L),小鼠卵母细胞在体外成熟培养12 h后,检测卵母细胞第一极排出情况、卵母细胞胞质内活性氧(ROS)和谷胱甘肽(GSH)含量,并进行体外受精(IVF);体外受精后6 h统计受精率,24 h统计卵裂率,96 h统计囊胚率。结果显示,与对照组相比,10、15 μmol/L藏红花素显著提高了卵母细胞第一极体排出率(P<0.05);当藏红花素浓度继续增加时,卵母细胞的第一极体排出率下降,30 μmol/L藏红花素显著降低了卵母细胞第一极体排出率(P<0.05);5、10、15 μmol/L藏红花素均显著降低了卵母细胞ROS含量(P<0.05),10、15 μmol/L藏红花素均显著提高了卵母细胞GSH含量(P<0.05)。与对照组相比,10 μmol/L藏红花素组受精率、卵裂率、囊胚率差异均不显著(P>0.05),15、20、25、30 μmol/L藏红花素均显著降低受精率和囊胚率(P<0.05),对卵裂率影响不显著(P>0.05)。结果表明,在小鼠卵母细胞体外成熟培养液中添加10 μmol/L藏红花素可以显著增加第一极体排出率,显著降低卵母细胞ROS含量、提高卵母细胞GSH含量,但对受精后的胚胎发育无显著影响。 相似文献

本试验主要比较了离子霉素、电脉冲两种方法激活牛、羊体外成熟卵母细胞的效率。两种激活方法中。牛胚胎卵裂率无显著差异(90.61%对94.40%,P〉0.05),而离子霉素激活胚胎的囊胚发育率极显著高于电激活方法(12.3%对2.4%,P〈0.01)。两种方法对羊胚胎的研究中,羊胚胎卵裂率无显著差异(72.4%对77.4%,P〉0.05)。但是离子霉素激活胚胎的囊胚发育率显著高于电激活方法(3.67%对10.40%,P〈0.05)。本试验中还比较了用化学激活法(离子霉素)激活牛体外成熟卵母细胞后,用SOFaa体系培养,换液与不换液对孤雌激活胚胎体外发育的影响。结果表明:在第4天不换液的胚胎卵裂率和囊胚率极显著高于换液的胚胎(11.64%对3.49%。P〈0.01)。 相似文献

研究季节对延边黄牛卵巢质量和卵母细胞的数量、质量、体外成熟以及核移植重组胚发育的影响。持续1年采集延边黄牛卵巢,分级卵巢,抽取卵巢表面直径2~8mm的卵泡内的卵母细胞,收集后进行体外成熟和重组胚的体外培养。结果表明,1级卵巢的比例在春冬季时明显高于秋季(P〈0.05),2级卵巢的比例在春夏季高于冬季(P〈0.05),秋季的3级卵巢比例最高,与其他季节有显著差异(P〈0.05);在秋冬季,平均每个卵巢拣出卵母细胞数高于春夏季(P〈0.05),成熟率上,秋季显著高于春夏季(P〈0.05),但与冬季并无差异且其他各组间无显著差异(P〉0.05);卵裂率上,秋季与春季有差异(P〈0.05),但与其他季节无显著差异(P〉0.05),囊胚率上,各组之间均无显著差异(P〉0.05)。通过研究发现,季节对延边黄牛卵母细胞的可用回收数量及其质量都会产生影响且秋季为延边黄牛体细胞核移植实验最适合季节。 相似文献

对不同的培养液(M199、mDPBS、TYH)和培养液添加物对小鼠C级卵母细胞体外发育和成熟的影响进行了研究。结果表明,mDPBS培养液的培养效果(GVBD:19.8%,PBI:12.3%)好于(P〈0.05)M199培养液(GVBD:13.3%,PBI:8.9%)和TYH培养液(GVBD:12.3%,PBI:7.4%);mDPBS中添加丙酮酸钠和FCS(GVBD:100%,PBI:86.3%)明显优于(P〈0.01)单纯添加丙酮酸钠(GVBD:42.9%,PBI:23.8%),也明显优于(P〈0.01)添加丙酮酸钠+BSA(GVBD:63.2%,PBI:53.8%);就A、B和C级3种卵母细胞相比,C级卵母细胞的成熟发育最佳,GVBD的发生率为100.0%,PBI的排出率为86.2%。 相似文献

研究卵母细胞发育程度和不同营养因子对小鼠卵母细胞体外成熟及体外受精率的影响。小鼠卵母细胞分别在含有FSH、BSA和胰岛素的培养液中体外成熟 ,在Whitten氏液中体外受精 ,比较体外成熟率、体外受精率。卵丘卵母细胞 (COC)较裸卵 (DO)的体外成熟率、体外受精率都低。FSH和胰岛素均能提高小鼠卵母细胞的体外成熟率、体外受精率。BSA可以降低小鼠卵母细胞体外受精率。GV期卵母细胞的体外受精率显著低于体外培养的排出第一极体的卵母细胞。 (P2 -cell <0 0 5 ,P受精 <0 0 5)。 相似文献

为了研究不同培养时间对家猫卵母细胞核成熟的影响,以M 199为基础培养液,并添加FSH、LH,分别培养24、28、32、36和43 h后,观察第一极体排出,然后用地衣红染色,将排出第一极体和染色体处于MⅡ期作为卵母细胞成熟的参考判断标志。结果表明,家猫卵母细胞随着培养时间的延长其成熟率也随之增加。成熟培养43 h,极体排出率极显著高于其他各组(P<0.01),达到66.89%;而24 h组极体排出率为31.51%,极显著低于其他各组(P<0.01);28、32、36 h各组间差异不显著(P>0.05)。经地衣红染色后,到达MⅡ期的卵母细胞比例与极体排出率趋势相同。43 h组退化率极显著高于其他各组(P<0.01),达到22.30%。因此,综合考虑成熟率和退化率两项指标,28~36 h为家猫卵母细胞体外核成熟的最佳培养时间。 相似文献

体外成熟的绵羊卵母细胞,经5 μmol/L A23187 5 min和2 mmol/L 6-DMAP 4 h激活后,分别在SOFaa、M-199两种培养液中进行培养,试验比较了SOFaa、M-199液分别添加BSA和不同浓度(10%、20%)胎牛血清对绵羊孤雌生殖胚体外发育的影响。M-199+FCS组的卵裂率显著低于SOFaa+BSA和SOFaa+FCS组(P<0.05),SOFaa+BSA组的囊胚率显著低于M-199+BSA、SOFaa+FCS、M-199+FCS三组(P<0.05);SOFaa+10% FCS与SOFaa+20% FCS组的卵裂率及囊胚率均无显著差异,M-199+10% FCS和M-199+20% FCS组的卵裂率及囊胚率间均无显著差异。结果表明:SOFaa比M-199更适合绵羊孤雌生殖胚体外发育,此外添加10%的FCS即可达到较好的培养效果。 相似文献

以国际标准强毒R株人工感染非免疫产蛋鸡,定时扑杀,分别从鼻窦、眶下孔、气管、肺、气囊、卵巢和输卵管分离MG,并收集感染鸡所产蛋分离MG。结果表明,人工感染48小时后上、下呼吸道及肺已被全面感染,96小时气囊已被感染,120小时输卵管已能分离到MG,卵巢始终分离不到MG。人工感染鸡自144小时便能在其所产蛋中分离出MG。药物治疗能在72小时内消除感染,油乳剂苗则需24天后逐渐降低蛋内MG分离率,药物卵内注射、种蛋药浴、高温处理均能杀死卵内MG,但以研制的种蛋浸泡剂药浴效果为最好。 相似文献

REASONS FOR PERFORMING STUDY: Centesis of the bicipital bursa using an 8.9 cm long spinal needle has been reported but the alternative of employing a 3.8 cm long hypodermic needle requires validation. OBJECTIVE: To compare the efficacy of 2 different methods of centesis of the bicipital bursa and to evaluate the usefulness of ultrasonographic imaging to determine the location of solution administered when centesis of the bursa is attempted. METHODS: For Trial 1, 6 clinicians, who had no previous experience of centesis of the bicipital bursa, attempted to inject a solution composed of an aqueous radiopaque contrast medium and physiological saline solution (PSS) into the bicipital bursae of 2/12 horses using the previously described distal approach to inject one bursa and a proximal approach to inject the contralateral bursa. The bicipital tendon and bursa were examined ultrasonographically before and after injection; and both shoulders were examined radiographically to identify the location of the medium. In Trial 2, another 6 clinicians, also with no previous experience of centesis, repeated Trial 1, using 6 horses, but the radiopaque contrast medium was mixed with air instead of PSS. RESULTS: Accuracy of centesis using the proximal approach was 39% and that of the distal approach 28%. Ultrasonographic examination of the shoulder allowed the location of solution and air to be accurately predicted in all 12 shoulders examined. CONCLUSIONS: Clinicians who have had no previous experience performing centesis of the bicipital bursa are unlikely to be successful in centesis using either approach. Radiographic examination after injecting a radiopaque contrast medium may be necessary to assess the success of centesis especially if bursal fluid is not obtained during centesis. Injecting air along with the radiopaque contrast medium provides more accurate ultrasonographic confirmation of centesis and better radiographic definition than does injection without air. 相似文献

Effects of size of ingestively masticated fragments of plant tissues on kinetics of digestion of NDF
Ingestively masticated fragments were collected and sized via sieving. Different sizes of esophageal masticate and ruminal digesta fragments, and ground fragments of larger masticated pieces were incubated in vitro, and undigested NDF remaining at intervals of up to 168 h of incubation was determined. The ruminal age-dependent time delay (tau) for onset of digestion of NDF was positively correlated (P < 0.004) with the mean sieve aperture estimated to retain 50% of the fragments between successive sieve apertures (MRA). Degradation rate of potentially degradable NDF (PDF) and level of indigestible NDF were not related (P > 0.10) to MRA of masticated and ground fragments. Estimates of tau were positively related to MRA, with slopes of bermudagrass < corn silage < ruminal fragments of corn silage. It was concluded that fragment size-, and consequently, ruminal age-dependent onset of PDF degradation of a mixture of different fragment sizes results in an age-dependent rate of degradation of the more rapidly degrading of two subentities of PDF. Models are proposed that assume a tau before onset of simultaneous degradation of PDF from two pools characterized as having gamma-modeled age-dependency and age-constant rates. The ruminal age-dependent pool seems to be associated with the faster-degrading pool, and its rate parameter increases with range in MRA in the population of fragments. Conceptually, the ruminal age-dependent rate parameter for PDF degradation seems to represent a composite of several effects: 1) effects of the size-dependent tau; 2) range in MRA of the population of ingestively masticated fragments; and 3) subentities of PDF that degrade via more rapid age-dependent rates compared with subentities of PDF that degrade via age-constant rates. The estimated fractional rates of ruminative comminution of ingestively masticated fragments (0.060 to 0.075/h) were of a magnitude similar to the mean fractional rates of PDF digestion (0.030 to 0.085/h), which implies that ruminative comminution may be first-limiting to fractional rate of PDF digestion. The in vivo roles of ingestive and ruminative mastication of fragments on PDF degradation must be considered in any kinetic system for estimating PDF digestion in the rumen. These results and others in the literature suggest that the rate of surface area exposure rather than intrinsic chemical attributes of PDF may be first-limiting to degradation rate of PDF in vivo. 相似文献
