从江苏徐州地区分离到的以引起樱桃谷鸭产蛋下降和死亡为特征的1株病毒,命名为XZ株。对该病毒进行电镜观察、血凝试验、ELD50测定、RT-PCR扩增特异性目的基因、序列比对分析和动物回归试验。结果显示,分离毒株能致死鸭胚和鸡胚,电镜下观察到球形病毒粒子,不具有血凝性,对病料和接毒鸭胚尿囊液进行RT-PCR,均可扩增出基因片段,其核苷酸序列与坦布苏病毒奉贤株的相似性最高,为98.7%,与其他坦布苏病毒的毒株也具较高同源性,为86%~98%。用鸭胚分离毒株接种健康产蛋鸭,能复制出同样的疾病。结果表明分离病毒为鸭黄病毒属的坦布苏病毒。  相似文献

本试验通过病理解剖、组织学检查、电镜观察及RT-PCR等方法对一起鸭产蛋下降性疾病进行初步研究。解剖病鸭发现,脾脏肿大,卵泡膜出血、变性;组织学检查结果表明,病鸭肝细胞呈现严重的脂肪变性,脾脏淋巴细胞数量减少;病料细菌分离结果为阴性;黄病毒RT-PCR检测阳性;电镜下可观察到直径为40~60 nm的病毒粒子;参照黄病毒属NS5基因设计合成引物,扩增得到360 bp的目的片段;遗传进化分析表明:该核酸序列与黄病毒科黄病毒属恩塔亚病毒群(Ntaya virus group)的巴格扎病毒(Bagaza virus)的NS5核酸序列同源性最高。  相似文献

鸭传染性产蛋减少症(暂定名)的病原分离初报   总被引:3,自引:0,他引:3  
本文对2010年在成年蛋鸭中流行的以产蛋下降为特征的未知疫病病原进行了初步鉴定。病原能在鸭胚、鸡胚和细胞上复制增殖,对鸭胚、鸡胚的致死率达100%。鸭胚病毒分离物不具有血凝性,在透射电镜下可观察到直径为50~100 nm、有囊膜带突起的病毒粒子。病毒对酸敏感、能耐受pH9的碱性环境,50℃1 h可使病毒失去感染能力。SPF鸡人工感染试验结果显示,感染鸡生长缓慢、致死亡率低、能回收病毒。说明引起鸭产蛋量下降的疫病病原是病毒性的,并导致鸡感染发病,我们将此病暂称为鸭传染性产蛋减少症。  相似文献

鸭出血性卵巢炎的初步研究   总被引:27,自引:1,他引:26  
2010年6月以来,我国部分地区所饲养的种鸭和蛋鸭发生一种疾病,以突发产蛋急剧下降为特点,根据病变,暂将该病称为鸭出血性卵巢炎(duck hemorrhagic ovaritis,DHO)。从不同地区的发病鸭群采集34份样品,取15份样品进行病毒分离,获得10个鸡胚分离物和5个鸭胚分离物。RT-PCR检测结果表明,15个分离物均为禽流感阴性,14个分离物为新城疫阴性。选分离株YY5株进行了电镜观察和PCR排查,结果表明,该毒株的毒粒直径为40~50 nm,呈鸭瘟病毒、水禽细小病毒、鸭呼肠孤病毒、鸭甲肝病毒、鸭星状病毒、鸭圆环病毒、鸭冠状病毒、鸡传染性支气管炎病毒PCR阴性,但为黄病毒RT-PCR阳性。基于黄病毒NS1序列合成引物,用RT-PCR检测15株病毒分离物和其余19份临床样品,黄病毒阳性率为82.4%。用YY5株的221-nt NS1序列进行分析的结果显示,YY5株与黄病毒科黄病毒属恩塔亚病毒群(Ntaya virus group)的巴格扎病毒(Bagaza virus)具有相近的遗传进化关系,但遗传距离介于病毒种的水平。用一株鸭胚分离株感染91日龄临近开产的麻鸭和232日龄的产蛋麻鸭,可复制出卵泡膜出血的病变。结果表明,从DHO自然病例分离到一种新的黄病毒,暂称为鸭黄病毒(duck flavivirus,DFV),DFV可能与DHO有关。  相似文献

为探明鸭坦布苏病毒对产蛋鸡的致病性和不同感染途径对其产蛋性能的影响,分别选用罗曼蛋鸡、海兰蛋鸡和淮南麻黄鸡3个品种进行人工接种病毒试验。结果表明,3个品种产蛋鸡接种病毒后均出现产蛋下降,产蛋率最高下降80%以上,除产蛋和早期采食量下降及后期严重脱羽外,感染鸡无其他临床症状,产蛋恢复的速度和程度不同品种间存在一定差异,感染后无鸡只死亡;滴鼻组和同居组鸡群产蛋下降和恢复的程度低于肌肉注射组,重复感染组未发生明显产蛋下降;剖检病变可见一致的卵巢组织出血、变性、坏死和萎缩,其他器官无肉眼可见病变;从感染鸡卵巢组织中可检测到鸭坦布苏病毒核酸。综上所述,鸭坦布苏病毒对不同品种产蛋鸡均具有致病性,主要引发产蛋下降,病毒在鸡群中的传播可能不仅限于吸血昆虫,空气传播感染也是其重要的传播途径;感染康复鸡只对再次感染具有一定的抵抗力。研究结果为产蛋鸡不明原因产蛋下降提供了新的诊断方向,为坦布苏病毒病的有效防控发出了预警信号。  相似文献

从广东某番鸭种鸭场全场有明显呼吸道症状和产蛋下降,而无其他典型鸭副黏病毒病症状的种番鸭群中分离到1株副黏病毒(命名为DP0690)。经过鉴定,该分离株具有血凝性,可被新城疫病毒(NDV)标准阳性血清所抑制,不能被禽流感病毒(AIVH5与H9亚型)阳性血清和减蛋综合征病毒(EDSV)标准阳性血清抑制;RT-PCR检测为NDV阳性;对F基因进行部分测序和分析表明F基因的裂解位点序列为112R-R-Q-R-R-F117;动物回归实验表明:该分离毒株对5日龄SPF雏鸡和雏番鸭能100%致死;对开产母鸭能复制出相同的病例,表明该病毒属强毒力毒株。  相似文献

致麻鸭产蛋下降的副粘病毒的分离和鉴定   总被引:16,自引:1,他引:16  
从浙江省某麻鸭养殖基地产蛋锐减的病鸭生殖器官分离到1株致产蛋下降、不致鸭死亡的病毒株(YH99V)。该病毒易感蛋鸭,经SPF鸡胚传至第9代时致病性突然增强,第11代出现对鸡红细胞的血凝特性,通常在42~80h致死SPF鸡胚,EF15EID50为10^1.8。经磷钨酸负染电镜观察,YH99V粒子呈现圆形、杆状形、葫芦状形等多形态,直径70~400nm不等,病毒外表有囊膜,囊膜外层有排列整齐的纤突。病毒粒子和包涵体位于细胞浆内。病毒抵抗力由弱到强依次为:0.2%甲醛、24h-56℃、45min→紫外线、1h→乙醚≈氯仿≈37℃、16h—pH9→pH5→1%Try。对人眼结膜易感,鸭眼结膜不易感。接种传代细胞BHK-21、IBRS-2均能引起细胞病变。YH99V株与同样引起鸭产蛋下降的AIV、鸭源EDSV无抗原相关性,而能被禽副粘病毒Ⅰ型阳性血清中和。根据试验结果,初步将YH99V判定为鸭副粘病毒。  相似文献

The health of oviduct circulation is very important for normal egg production. The present work describes the histology and histochemistry of vessels of different types on the oviduct of quail. Arteria, venae and lymphatics of the oviduct were described clearly. In addition, special vascular elements were observed presenting circular, longitudinal and oblique smooth muscle fibres on the blood vessel wall. Glomus cells were observed at typical glomus vessels, atypical glomus vessels and on the wall of some of arteria and arteriolae. Also, spirally oriented blood vessels were observed. Dark brown pigment was detected in the wall of arteria, venae, lymphatics and the special types of blood vessels.  相似文献

The effects of two lathyrogens (beta-aminopropionitrile and semicarbazide) on the reproductive system of the laying hen were compared by measuring the changes in the ovary and oviduct associated with a diminished egg production rate. The laying hens were fed diets containing either beta-aminopropionitrile, the toxic constituent of certain legume seeds in the genus Lathyrus or semicarbazide to induce lathyrogenic effects in which reduced egg production was one of the toxic effects. The gross examination of the reproductive system of the hens revealed that ovarian activity was greatly impaired. The reduced egg production after feeding beta-aminopropionitrile was due to the reduced growth rate of the ovary, and in the case of semicarbazide, to the atresia of large yolky follicles.  相似文献

The effect of a developing egg on the staining properties of the luminal epithelium of the isthmus and shell gland of the oviduct of the hen Gallus domesticus is described, with special reference to the red region. Using histochemical methods for the demonstration of complex mucopolysaccharides, the staining reactions in the surface epithelium adjacent to an egg were found to be similar to those seen in an empty oviduct. With the empirical methods for intracellular calcium, a staining reaction was seen in cells which were adjacent to an egg. There was no reaction in an empty oviduct. If an egg lay between the red region and the shell gland, methods for the demonstration of acid mucopolysaccharides or intracellular calcium showed a more pronounced reaction in an area of surface cells at the neck of the shell gland. The control of Newcastle disease (ND) in South Africa has proved difficult since 2002 following the introduction of lineage 5d/VIId Newcastle disease virus (NDV) strain ("goose paramyxovirus" - GPMV) to which commercially available ND vaccines appeared less effective. Most of the ND infections, even in fully vaccinated hens were characterized consistently by a drop in egg production. In this study, commercial and SPF hens-in-lay were vaccinated with La Sota vaccine and challenged with a GPMV isolate. Immunohistochemical labeling was used to determine the distribution of viral antigen in the oviduct of the hens. Following reports that cloacal vaccination offered better protection against egg production losses than the oro-nasal route, the efficacy of cloacal and ocular routes of vaccination against challenge were compared. Results showed that La Sota vaccine offered birds 100% protection against the virulent ND (GPMV) virus challenge from clinical disease and death, but not against infection and replication of the GPMV, as birds showed varying degrees of macropathology. Histopathology of the oviduct of infected birds revealed multifocal lymphocytic inflammation in the interstitium as well as mild glandular ectasia and mild edema. Finely granular NDV-specific immunolabeling was demonstrated in the cytoplasm of epithelial cells and mononuclear (lymphohistiocytic) cells in the interstitium of the oviduct. Both vaccine and virulent GPMV showed greatest tropism for the uterus (versus the magnum and isthmus). There was no clear difference in the protection of the oviduct and in the distribution of oviductal GPMV antigens between the two routes of vaccination.  相似文献

A 6-year-old cockatiel was diagnosed with egg binding based on clinical signs, physical examination findings, radiographs, and lack of oviposition. Medical management failed to induce oviposition, and surgical salpingotomy was used to remove the egg from the oviduct. Sterile egg yolk peritonitis was diagnosed based on histology. Postoperative management with leuprolide acetate and husbandary changes successfully prevented egg production for at least 2.5 months after the event.  相似文献

The objective of this study was to isolate and identify a hypothetical Chlamydiaceae pathogen from laying hens with an oviduct cyst, and to characterize its potential causal relation with decreased egg production. Our clinical survey showed that cystic oviducts were prevalent at rates of 10% and 15.1% in breeder and commercial hen flocks, respectively. Chlamydial antigens were detected in 20 of 50 pharyngeal swabs (40%) and in 17 of 20 oviduct tissues (85%) using enzyme-linked immunosorbent assay (ELISA) antigen detection kits. The isolated pathogen was identified as Chlamydophila psittaci via complement fixation test, PCE-ELISA, and immunofluorescence assay. Avian influenza virus, Newcastle disease virus, and infectious bronchitis virus were excluded after oviduct tissues were inoculated onto the chorioallantoic membrane of embryonating eggs. The nucleotide sequence of the omp1 gene (accession no. EF202608) from the isolate was similar to that of C. psittaci avian type C (accession no. L25436). Typical cystic oviducts were observed in specific-pathogen-free hens inoculated intraperitoneally with the isolate. The high presence of chlamydial antigen is consistent with the cystic oviducts and poor egg production. We conclude that the isolated C psittaci is most likely associated with cystic oviducts in laying hens.  相似文献

Studies were made on the distribution of lymphoid tissues and immunoglobulin (Ig: IgA, IgG and IgM)-containing cells (cIg: cIgA, cIgG and cIgM) and the localization of immunoglobulins (Igs) in the oviducal walls of laying hens. Lymphocyte accumulations were occasionally observed, located mainly in the middle infundibulum and in the regions from the isthmus to the vagina. The number of cIgG significantly predominated over that of cIgA or cIgM in the mucosal connective tissue of the magnum and the isthmus. In contrast, in the regions other than the magnum and the isthmus, these three types of cIg were fewer in number. Igs were localized in some superficial epithelial cells (SECs) and glandular cells (GlCs) of the oviduct. Many IgG-containing SECs were found in the infundibulum, the isthmus, and the cranial and major uterus. IgA- or IgM-containing SECs were rare throughout the oviduct. Three types of Ig-containing GlCs were numerously found in the magnum, though lymphocyte accumulations were scarce there. In the isthmus, many IgG-containing GlCs were found, while IgA- or IgM-containing GlCs were rarely observed. Ig-containing GlCs in the magnum were considerably decreased in number after the egg passage. The results suggest that the maternal Igs are transferred to the egg mainly through GlCs in the magnum of the chicken oviduct, and that the oviducal lymphoid tissues have little relationship to the passive immunity.  相似文献

The efficacy of the albumen test for infectious avian leukosis virus (ALV) was examined in detecting congenitally transmitting hens. Seventy-three White Leghorn non-viremic hens with antibody to ALV were used. Eleven of the hens shed infectious ALV into their egg albumen, whereas only 7 of the 11 ALV-positive hens shed ALV antigens. The egg albumen test for infectious ALV was shown to be more effective in detecting the congenitally transmitting hens than that for ALV antigens. Then, twenty of the 62 hens which shed no infectious ALV into the albumen were studied for transmission of ALV to their embryos and for discharging ALV into the oviduct and vagina. Six of the 50 embryos from 4 hens were found to be infected with ALV but all of the 227 embryos from remaining 16 hens were free from the infection. Discharge of the virus into the oviduct and vagina was found both in the 4 transmitting hens and in 6 of the 16 non-transmitting hens. These results suggest that the hens discharging ALV into the oviduct, even though they do not shed ALV into egg albumen, may transmit the virus sporadically to their embryos.  相似文献

The effect of a phyto-oestrogen, daidzein, on the laying performance of Shaoxing female ducks was examined in three experiments performed at different stages of the egg production cycle. Egg-laying rate, egg weight, egg composition, feed conversion ratio, hatchability characteristics of eggs and body weight, ovary and oviduct weight, as well as changes in serum concentrations of T3, T4 and E2 were recorded as response criteria. In the first experiment, 68 young ducks, 100 d of age, were given a basal diet (maize-soybean meal) with or without 3 mg of daidzein/kg diet for 42 d. Daidzein did not affect the onset of lay but apparently decreased egg-laying rate and mean egg weight as well as the feed conversion ratio. In the second experiment, 240 breeding ducks, 402 d of age, were allotted at random to three groups and given the basal diet containing daidzein at 0 (control), 3 (Da1) and 5mg/kg (Da2) for 35d. Egg-laying rate, mean egg weight and feed conversion ratio increased in both Da1 and Da2 groups. However, an adverse effect of daidzein on fertility and hatchability was observed. In the third experiment, 320 breeding ducks, 415 d of age, were fed on the basal diet with or without 5mg of daidzein/kg diet for 63 d. Egg-laying rate increased by 7.7%, average egg weight tended to increase, whereas yolk/albumen ratio decreased. Daidzein-treated ducks had higher body weight and oviduct weight compared with their controls. Elevated plasma T4 and E2 concentrations accompanied these phenotypic changes, but serum T3 was not affected. It is suggested that daidzein exerts divergent effects on the egg-laying performance of Shaoxing ducks under different physiological conditions and this action is dose-dependent. The changes in circulating E2 imply possible participation of endogenous oestrogen in the mechanism of daidzein action.  相似文献
