为使鸡源志贺氏菌IpaC蛋白保持天然的生物学活性,在毕赤酵母表达系统中实现分泌表达,进一步研究IpaC蛋白的生物学功能,本研究在不改变IpaC蛋白氨基酸序列的情况下,根据酵母密码子偏爱性优化合成IpaC#基因,利用真核表达载体p GAPZA和8种真核信号肽,分别构建真核表达载体pGAPZA-Inu/Inv/Kil/Lys/Mat/Amy/Glu/Ser-IpaC#,然后将线性化的重组质粒电转化至毕赤酵母X-33中,对阳性转化子进行表达,并用SDS-PAGE及Western-blot检测其表达产物。结果表明:在8种重组菌表达产物的菌体沉淀中检测到大小约为60 ku的目的蛋白。说明鸡源志贺氏菌IpaC基因成功实现了在毕赤酵母的胞内表达。  相似文献

为使鸡IL-18成熟蛋白基因在真核系统中高效表达,对其进行定点突变,将酵母低频密码子突变为偏嗜性密码子,构建了鸡IL-18成熟蛋白变构基因真核重组表达载体pPICZαA-MChIL18*,并将其转化入P.pastoris X-33.重组菌株X-33/pPICZαA-MChIL18*经甲醇诱导后,表达产物的上清液经SDS-PAGE检测,结果在约23 000处有目的条带,与预期的相对分子质量一致,占总蛋白的45%,对其进行Western-blot检测,结果与兔抗鸡IL-18多克隆抗体发生特异性免疫反应.结果表明,鸡IL-18成熟蛋白变构基因在毕赤酵母中成功的进行了表达.  相似文献

构建猪传染性胸膜肺炎放线杆菌(APP)apxIA基因的真核表达载体pPICZαA/apxI,并在毕赤酵母GS115中进行表达。SDS-PAGE显示仅在浓缩40倍的上清中检测得到表达产物,同时经RT-PCR可检测到重组酵母中编码目的基因成熟肽的mRNA,经分析发现目的序列AT含量高达62%,其中含49个毕赤酵母稀有密码子,占15.4%。证实ApxIA在毕赤酵母中为低水平表达。  相似文献

根据毕赤酵母(Pichia pastoris)密码子的偏嗜性,通过SOEing法合成改造后的Cryptonin基因片段。基因克隆到pPICZα-C质粒中,获得分泌型重组酵母表达载体pPICZα-C-Cryptonin。pPICZα-C-Cryptonin通过限制性内切酶SacⅠ酶切线性化后,经电穿孔法转入毕赤酵母细胞X-33内。PCR检测Cryptonin基因与毕赤酵母染色体稳定结合,Zeocin抗性筛选,得到高拷贝转化子。在AOX(醇氧化酶)启动子调控下,Cryptonin蛋白获得分泌表达,其相对分子质量约为2700。表达产物耐热性强,在100℃条件下10min内抗菌活性不变,煮沸30min以上仍具有活性。琼脂糖孔穴扩散法检测显示表达产物对革兰氏阳性菌和革兰氏阴性菌都具有很好的抑菌活性。  相似文献

将荣昌猪IFN-ω基因亚克隆到酵母表达载体pPICZαB后,氯化锂法转化毕赤酵母菌株X-33,通过酶切、PCR鉴定表明成功构建了表达荣昌猪IFN-ω基因的酵母基因工程菌。经甲醇诱导后,通过SDS-PAGE和Western blotting检测,表明荣昌猪IFN-ω基因在毕赤酵母中实现了高效表达。表达产物在MDBK细胞上抗VSV的比活性为1.21×106U/mg,表明具有抗病毒活性。  相似文献

用PCR方法从胸膜肺炎放线杆菌(APP)血清1型参考株APP259的基因组DNA中,扩增了约954bp的apxⅠA基因片段,分别构建了大肠杆菌表达载体pGEX-4T-1/apxⅠA和毕赤酵母表达载体pPICZαA/apxⅠA。对目的基因密码子偏好性进行了分析,发现其中有4个大肠杆菌稀有密码子,占1.2%;有49个毕赤酵母稀有密码子,占15.4%。利用所构建的大肠杆菌表达载体pGEX-4T-1/apxⅠA和毕赤酵母表达载体pPICZαA/apxⅠA分别进行表达。结果显示,目的基因在大肠杆菌中能以包涵体形式进行高效表达,其融合蛋白占菌体总蛋白的32%;在毕赤酵母中则以较低水平分泌表达至培养基的上清液中,40倍浓缩后方能检测到少量目的蛋白。证实,目的基因序列中的密码子偏好性影响其在大肠杆菌和毕赤酵母中的表达。  相似文献

为进一步研究IL-2对疫苗的免疫增强作用,本研究经PCR扩增分别获得鸡IL-2和新城疫病毒(NDV)HN基因片段,应用分子克隆技术获得含有融合基因IL2-HN的真核表达载体pPICZαA-IL2-HN,重组质粒pPICZαA-IL2-HN线性化后整合入毕赤酵母X-33染色体上,在甲醇诱导下表达融合蛋白,再进行SDS-PAGE、Western-blot检测。SDS-PAGE分析结果表明目的蛋白为分泌性表达,分子量约为100 ku,Western-blot结果表明目的蛋白具良好的免疫反应性。  相似文献

利用毕赤酵母系统表达O型口蹄疫病毒VP1基因,并对表达的蛋白进行免疫原性鉴定。将VP1基因克隆入毕赤酵母分泌性表达载体pPICZα-C中,获得重组表达载体pPICZαC-VP1。将重组质粒pPICZαC-VP1线性化后,用电穿孔法转入酵母菌X-33,用Zeocin+YPDS平板筛选重组子,经甲醇诱导表达后,SDS-PAGE和Western blot分析表达产物,并用酵母表达的VP1蛋白免疫小鼠,然后通过ELISA检测抗体水平。结果表明,酵母所表达的VP1蛋白能诱导小鼠产生特异性体液免疫应答。  相似文献

将猪α干扰素(PoIFN-α)基因克隆入毕赤酵母分泌型表达载体pPICZαA中,电转化毕赤酵母KM71H菌株,抗生素ZeocinTM浓度梯度筛选高抗性转化子,以PCR鉴定阳性重组菌株。筛选出的高拷贝重组菌株分别通过BMGY/BMMY培养基和YPG培养基,用甲醇诱导表达。SDS-PAGE和Westernblot印迹杂交结果证实,表达产物为重组PoIFN-α融合蛋白,经细胞毒性试验检测(WISH/VSV系统),表达产物的抗病毒活性为4.1×104IU/mL。  相似文献

采用高效液相色谱法测定癸氧喹酯干混悬剂的含量,在2-250μg/mL范围内,峰面积的常用对数与进样量浓度的常用对数呈良好的线性关系,R^2=1(n=5),平均回收率为99.24%~99.51%,RSD在0.05%~0.28%。此方法分析时间短,样品前处理简便、定量结果准确,重现性好,结果满意,为其质量控制提供了依据。  相似文献

REASONS FOR PERFORMING STUDY: Centesis of the bicipital bursa using an 8.9 cm long spinal needle has been reported but the alternative of employing a 3.8 cm long hypodermic needle requires validation. OBJECTIVE: To compare the efficacy of 2 different methods of centesis of the bicipital bursa and to evaluate the usefulness of ultrasonographic imaging to determine the location of solution administered when centesis of the bursa is attempted. METHODS: For Trial 1, 6 clinicians, who had no previous experience of centesis of the bicipital bursa, attempted to inject a solution composed of an aqueous radiopaque contrast medium and physiological saline solution (PSS) into the bicipital bursae of 2/12 horses using the previously described distal approach to inject one bursa and a proximal approach to inject the contralateral bursa. The bicipital tendon and bursa were examined ultrasonographically before and after injection; and both shoulders were examined radiographically to identify the location of the medium. In Trial 2, another 6 clinicians, also with no previous experience of centesis, repeated Trial 1, using 6 horses, but the radiopaque contrast medium was mixed with air instead of PSS. RESULTS: Accuracy of centesis using the proximal approach was 39% and that of the distal approach 28%. Ultrasonographic examination of the shoulder allowed the location of solution and air to be accurately predicted in all 12 shoulders examined. CONCLUSIONS: Clinicians who have had no previous experience performing centesis of the bicipital bursa are unlikely to be successful in centesis using either approach. Radiographic examination after injecting a radiopaque contrast medium may be necessary to assess the success of centesis especially if bursal fluid is not obtained during centesis. Injecting air along with the radiopaque contrast medium provides more accurate ultrasonographic confirmation of centesis and better radiographic definition than does injection without air.  相似文献

乳酸杆菌作为益生菌广泛用于人和动物。本文综述了乳酸杆菌改善宿主健康的机制。乳酸杆菌可通过产生抗菌物质如乳酸、过氧化氢、细菌素,或者通过竞争营养或肠道黏附位点来抑制致病菌;通过诱导黏附素的分泌或阻止细胞凋亡而增强肠道的屏障功能,从而保护肠道。文章重点讨论了乳酸杆菌表面成分(表面蛋白、脂磷壁酸和肽聚糖)与肠道受体(C型凝集素受体、Toll样受体和 Nod样受体),阐述了他们结合后启动免疫调节信号,调控肠道免疫功能以发挥改善健康作用的机制。  相似文献

Ingestively masticated fragments were collected and sized via sieving. Different sizes of esophageal masticate and ruminal digesta fragments, and ground fragments of larger masticated pieces were incubated in vitro, and undigested NDF remaining at intervals of up to 168 h of incubation was determined. The ruminal age-dependent time delay (tau) for onset of digestion of NDF was positively correlated (P < 0.004) with the mean sieve aperture estimated to retain 50% of the fragments between successive sieve apertures (MRA). Degradation rate of potentially degradable NDF (PDF) and level of indigestible NDF were not related (P > 0.10) to MRA of masticated and ground fragments. Estimates of tau were positively related to MRA, with slopes of bermudagrass < corn silage < ruminal fragments of corn silage. It was concluded that fragment size-, and consequently, ruminal age-dependent onset of PDF degradation of a mixture of different fragment sizes results in an age-dependent rate of degradation of the more rapidly degrading of two subentities of PDF. Models are proposed that assume a tau before onset of simultaneous degradation of PDF from two pools characterized as having gamma-modeled age-dependency and age-constant rates. The ruminal age-dependent pool seems to be associated with the faster-degrading pool, and its rate parameter increases with range in MRA in the population of fragments. Conceptually, the ruminal age-dependent rate parameter for PDF degradation seems to represent a composite of several effects: 1) effects of the size-dependent tau; 2) range in MRA of the population of ingestively masticated fragments; and 3) subentities of PDF that degrade via more rapid age-dependent rates compared with subentities of PDF that degrade via age-constant rates. The estimated fractional rates of ruminative comminution of ingestively masticated fragments (0.060 to 0.075/h) were of a magnitude similar to the mean fractional rates of PDF digestion (0.030 to 0.085/h), which implies that ruminative comminution may be first-limiting to fractional rate of PDF digestion. The in vivo roles of ingestive and ruminative mastication of fragments on PDF degradation must be considered in any kinetic system for estimating PDF digestion in the rumen. These results and others in the literature suggest that the rate of surface area exposure rather than intrinsic chemical attributes of PDF may be first-limiting to degradation rate of PDF in vivo.  相似文献

以国际标准强毒R株人工感染非免疫产蛋鸡,定时扑杀,分别从鼻窦、眶下孔、气管、肺、气囊、卵巢和输卵管分离MG,并收集感染鸡所产蛋分离MG。结果表明,人工感染48小时后上、下呼吸道及肺已被全面感染,96小时气囊已被感染,120小时输卵管已能分离到MG,卵巢始终分离不到MG。人工感染鸡自144小时便能在其所产蛋中分离出MG。药物治疗能在72小时内消除感染,油乳剂苗则需24天后逐渐降低蛋内MG分离率,药物卵内注射、种蛋药浴、高温处理均能杀死卵内MG,但以研制的种蛋浸泡剂药浴效果为最好。  相似文献
