Three different versions of the conventional PCR described by Bulman & Marshall ( 1997 ) for the identification of cysts and juveniles of Globodera pallida and G. rostochiensis were compared: the original Bulman & Marshall, Bulman & Marshall as described in EPPO PM 7/40 (2) and an in‐house modified version of the Dutch National Plant Protection Organization (NPPO‐NL). The versions differ from each other in thermocycler conditions and primer sequences. Two different polymerases (Invitrogen and Roche) were assessed using the different test versions, and performance criteria analytical sensitivity and analytical specificity were determined. Roche‐based reaction mixes had the highest amplicon yield and were used for further comparison of the different test versions. The different test versions performed equally well in terms of analytical specificity. No false positive or false negative results were observed. The test version NPPO‐NL proved to be the most sensitive test version with a limit of detection of 1 juvenile for both G. pallida and G. rostochiensis.  相似文献

Few plant parasitic nematodes are currently included in the European Union (EU) regulated pest lists. However, many plant parasitic nematodes not present in the EU are known to be damaging and present a risk to Europe. Therefore a study was performed to assess the risk of importing plant parasitic nematodes with the soil attached to plants for planting, with emphasis on plants in pots. Prior to the survey, a list of harmful plant parasitic nematodes not present within Europe was prepared per continent based on a literature review. This resulted in a Risk List of 26 species. Additionally, over a 3‐year period a survey was carried out on plants for planting imported to the Netherlands with adhering soil, focusing on all plant parasitic nematodes and with special attention to the nematodes on the Risk List. A total of 258 soil samples were studied, originating from 54 different plant species and 20 different countries. Despite the small number of samples, several quarantine nematodes and species from the Risk List were detected, including new nematode–plant combinations. This survey illustrates that plants with adhering soil can be a pathway for the introduction of listed nematodes, including those from the Risk List, to the EU/EPPO region. This information might encourage a more risk‐based approach to performing import inspections.  相似文献

ABSTRACT Hybridization between two meiotic parthenogenetic species of root-knot nematodes, Meloidogyne chitwoodi and M. fallax, was investigated in two different crossing experiments on tomato plants grown in sand. The first experiment was a controlled cross between the two species. The second experiment was a bulk mating in a 1:1 mixture of two isolates. The haploid chromosome number of the parental isolates was n = 18. Successful interspecific hybridization was obtained, and the resulting hybrids produced egg masses. In eggs, cell division was observed, but most of them were without clear differentiation and consequently were sterile. Hatched F(2) juveniles were small in number, not viable, and showed morphological distortions. In the progeny of the isolate mixture of the bulk mating experiment, parental-type females of the two isolates were present in equal numbers, and 10% of all females were nonviable hybrids. Similar ratios of parental-type and hybrid females were detected in roots of test plants grown in soil from a field sample that contained a mixture of M. chitwoodi and M. fallax populations. In the controlled cross experiment, isozyme electrophoresis of malate dehydrogenase was applied to distinguish the two species and their hybrids. In the bulk mating experiment, malate dehydrogenase, esterase, and glucose 6-phosphate dehydrogenase were used as markers, two by two simultaneously on the same individual females, providing conclusive evidence for the occurrence of hybrids. This is the first report on interspecific hybridization in Meloidogyne. The possible role of interspecific hybridization in species differentiation and interspecific exchange of genetic material within Meloidogyne is discussed.  相似文献

The rice leaf nematode, Aphelenchoides besseyi, was isolated in 2017 from rice seeds cultivated in Romania. Identification was based on morphology and confirmed with a real‐time PCR test. This paper provides the first report of A. besseyi in Romania.  相似文献
